丹参用于兔股骨头坏死的治疗
Establishment of a Hydrocortisone-Induced Femoral Head Necrosis Model in Rabbit and Its Treatment with Chinese Traditional Medicine Danshen Root
【摘要】 目的建立兔股骨头坏死动物模型并应用于股骨头坏死的治疗研究。方法用氢化可的松注射液按每次10mg/kg肌内注射,每周2次连续给药6周,第12周每组随机抽取2只解剖进行肉眼观察,数字摄片,光镜、电镜检查,并进行手术治疗。结果给氢化可的松6周后,第12周兔激素性股骨头坏死动物模型已建立。随后进行手术治疗,治疗组均比对照组股骨头损坏程度较轻,尤其第2治疗组用浸透丹参的同种异体骨植入填塞效果更好,采用Ridit分析,第2治疗组与第1治疗组、第3治疗组之间差异有显著性意义(P<0.05)。对照组呈进行性坏死。结论长期大量使用氢化可的松注射液可引起激素性股骨头坏死,采用浸透丹参的同种异体骨植入填塞,疗效较好,对临床治疗激素性股骨头坏死可能有积极的参考意义。
【Abstract】 Objective To establish a rabbit model of femoral head necrosis and to explore its treatment. Methods Hydrocortisone in a dose of 10 mg·kg~ -1 was administered by i.m. injection twice per week for six weeks. At the 12th week two rabbits were randomly sacrificed and tissue samples from the femoral head were taken for assessment by gross inspection, digital photography, optic and electron microscopy. Treatment by drilling and Chinese traditional medicine Danshen root (Radix Salviae Miltiorrhizae) was applied, respectively. Results After injecting hydrocortisone for six weeks, the rabbit model of aseptic femoral head necrosis was successfully established. All the histological and electron microscopic observations and Ridit test showed that the femoral head lesions were apparently less serious in the group treated by surgical drilling plus Danshen-soaked allogenous graft than that in the group treated by surgical drilling plus pure allogenous graft and in the group with surgical drilling alone, and the lesions were progressively worsening in the control group. Conclusions It is feasible to establish a femoral head necrosis model in rabbits by continuous injection of hydrocortisone in high dosage. A Chinese traditional medicine Danshen-soaked allogenous graft may be beneficial in the treatment for this disease and worthy of further studies.
