抗苗勒氏管激素对人黄素化颗粒细胞激素分泌的影响
Effect of anti-Müllerian hormone on hormone secretion in cultured human luteinized granulose cells
【摘要】 目的:探讨抗苗勒氏管激素(AMH)对人颗粒细胞激素产生分时段的影响。方法:采用人黄素化颗粒细胞原代培养,分组加入不同浓度的AMH,分时段测定细胞培养液中的雌二醇(E2)和孕酮(P)浓度。结果:细胞培养液中E2、P浓度均随培养时间延长而升高,升高幅度渐下降。随着AMH浓度的增加,颗粒细胞的E2和P分泌明显降低,AMH5μg/L组到20μg/L组间有剂量依赖性,AMH 50μg/L组与20μg/L组比较无显著差别。结论:AMH可以降低体外培养人黄素化颗粒细胞的雌、孕激素分泌,并有一定的剂量依赖性,提示AMH可以影响颗粒细胞的激素合成过程。
【Abstract】 AIM:To explore the effect of anti-Müllerian hormone(AMH)on hormone secretion of cultured human luteinized granulosa cells.METHODS:Human luteinized granulose cells were cultured with different concentrations of AMH.The levels of estrogen and progesterone in the supernatants were measured.RESULTS:The levels of estrogen and progesterone increased gradually and peaked at 72 h.The secretions of estrogen and progesterone were significantly inhibited by AMH with concentration dependence(5 μg/L to 20 μg/L).No significant difference was found in estrogen and progesterone levels in the presence of AMH at concentrations from 20 μg/L to 50 μg/L group.CONCLUSION:AMH decreases the secretion of estrogen and progesterone in cultured human luteinized granulose cells in a concentration dependent manner,suggesting that AMH negatively affect hormone secretion in granulose cells.
