脂质体转染对HSV-2多表位复合DNA疫苗诱导小鼠特异性免疫应答的影响

【摘要】 目的研究不同免疫方式对HSV-2多表位复合DNA疫苗诱导小鼠免疫应答的影响,探索新型HSV-2DNA疫苗的有效免疫方式。方法利用真核表达质粒pcDNA3.1构建HSV-2糖蛋白gD,gB,gC和早期表达蛋白ICP 27的多表位复合DNA疫苗HV-pcDNA3.1。C57/BL6小鼠分为脂质体包裹pcDNA3.1空载体质粒对照组、脂质体包裹HV-pcDNA3.1质粒和裸质粒HV-pcDNA3.1肌肉注射免疫三组。ELISA检测小鼠血清HSV-2特异性IgG,IL-2和IFN-γ,乳酸脱氢酶法检测杀伤性T淋巴细胞(CTL)活性。结果与裸质粒免疫组相比,脂质体包裹HV-pcDNA3.1免疫组小鼠血清中HSV-2特异性IgG和IFN-γ表达明显增加,CTL活性增强。结论脂质体包裹质粒比裸质粒肌肉注射方式免疫小鼠能显著提高HSV-2多表位复合DNA疫苗的免疫活性。

【Abstract】 Objective To characterize the specific immune responses induced by liposome-coated multigenic DNA vaccine of herpes simplex virus type 2(HSV-2) via intramuscular routes,to find an effective immunization approach for this novel DNA vaccine.Methods The multigenic vaccine HV-pcDNA3.1 was constructed by inserting the glycoprotein D(gD),gB,gC and infectious cell protein 27(ICP 27)into the eukaryotic expression vector pcDNA3.1.The C57/BL6 mice were inoculated intramuscularly by the liposome-coated pcDNA3.1 and liposome-coated HVpcDNA3.1 and the naked HV-pcDNA3.1 respectively.The levels of specific IgG,IL-2 and IFN-γ in serum were determined by using ELISA.The function of CTL was detected by LDH assay.Results The liposome-coated HV-pcDNA3.1 remarkably enhanced the levels of the specific IgG,IFN-γ and CTL when compared with naked HV-pcDNA3.1.Conclusion Inoculation subcutaneouly by the liposome-coated plasmid remarkably enhanced the immunological competence of multigenic DNA vaccine of HSV-2 when compared with naked plasmid.
