神经胶质瘤细胞对血管新生作用及其血管生成素-2表达的影响

【摘要】 目的:探讨人神经胶质瘤细胞对血管新生的影响及作用机制。方法:体外分离培养人神经胶质瘤细胞,用免疫细胞化学法检测不同级别的肿瘤细胞中血管生成素-2的表达,采用图像分析软件进行光密度(IOD)的测量;培养人脐带静脉内皮细胞并应用Matrigel建立血管形成的体外培养体系,检测肿瘤细胞对血管新生的影响。结果:血管生成素-2在神经胶质瘤细胞中的表达强度与瘤组织的分级呈正相关(P<0.05);肿瘤细胞诱导肿瘤血管形成呈浓度依赖效应(P<0.05)。结论:神经胶质瘤细胞可以诱导肿瘤血管新生,血管生成素-2在神经胶质瘤细胞中的表达增强与肿瘤血管新生过程相关。

【Abstract】 Objective: To discuss the influence of the role of human neurospongioma cells infacilitating human vascular endothelial cells and expression of neurospongioma angiogenin-2.Methods:HUVECs were cultured by modified Jaffe methods and induced to develop tubular structure by Matrigel.Thus set up a stable angiogenesis system and added the stimulating factor.Neurospongioma cells were fractional cultivated in vitro and the expression of Ang-2 protein was detected by immuncytochemical methods. Finally the date was analyzed by statistics.Results:HUVECs could be induced to form tubular structure by Matrigel in vitro. After treatment with the conditional medium of tumor cells, the number of tubules increased in a dose-dependent manner. The expression of Ang-2 increased in neurospongioma cells.Conclusion:Angiogenesis can be induced by neurospongioma cells;the expression of Ang-2 in neurospongioma upregulates and in positive correlation with grade of tumor.
