紫杉醇诱导人胃癌MGC803细胞凋亡作用研究

【摘要】 目的:研究胃癌发生过程中原位细胞凋亡活动以及紫杉醇对人胃癌细胞系MGC803诱导细胞凋亡的作用。方法:用脱氧核糖核酸末端转移酶介导的缺口末端标记法检测了胃癌MGC803细胞凋亡,用紫杉醇处理培养胃癌细胞MGC803、显微镜下进行观察并且进行琼脂糖电泳,流式细胞仪检测细胞周期分布和端粒体酶活性检测。结果:癌旁对照组原位凋亡细胞增多,紫杉醇处理胃癌细胞可以诱导细胞产生典型的细胞凋亡变化。DNA发生断裂,琼脂糖电泳产生梯形条带,细胞阻滞于G2/M期,端粒体酶活性下降。结论:在体内肿瘤细胞凋亡降低,细胞分裂可加快,癌旁对照中细胞凋亡增多,体外紫杉醇可以诱导胃癌细胞系MGC803凋亡。

【Abstract】 Objective:To investigate the action of insitu apoptosis in occurrence of gastric cancer and the biological effect of Paclitaxel on MGC803 cell line inducing apoptosis.Methods:The apoptosis in the gastric cancer detected with terminal deoxynucl neotidyl transferase mediated dUTP nick end labelling(TUNEL),the MGC803 cell line cultured in medium were treated with Paclitaxel and observed undermicroscope,and their DNA was assayed by gel electrophoresis,Flow cytometry analysis was used to observe the cell cycle,the effect measured telomerase.Results:In situ apoptosis cells in the cancer adjacent tissues inceased;treatment of gastric cancer with Paclitaxel could induce the cells to manifest a typical apoptosis morphology.Their DNA was fractured and a characteristic ladder pattern could be found using electrophoresis,which was associated with arrest of G2/M phase,and inhibited telomerase activity.Conclusion:In vivo the apoptosis of gastric cancer lower,maybe the cells divide quickly and then the cancers occur.In the cancer adjacent tissues,the apoptosis prick up,and in vitro Paclitaxel can induce the apoptosis of MGC803 cells.
