黄连解毒汤中三种成分在大鼠体内的药代动力学研究

【摘要】 目的:研究黄连解毒汤中盐酸小檗碱、黄芩苷、栀子苷在大鼠体内的药代动力学。方法:健康大鼠灌胃给予黄连解毒汤,收集不同时间的含药血浆,采用HPLC法测定大鼠血浆中盐酸小檗碱、黄芩苷、栀子苷三种成分的血药浓度,运用3P97药代动力学软件拟合房室模型,计算其药动学参数。结果:黄连解毒汤中盐酸小檗碱和黄芩苷在大鼠血浆中的药-时过程属于一室开放模型,栀子苷在大鼠血浆中的药-时过程属于二室开放模型。结论:建立了HPLC法测定黄连解毒汤中三种成分血药浓度的方法,为黄连解毒汤的合理用药提供了药代动力学数据,同时也为中药复方的药代动力学研究提供了可借鉴的方法。

【Abstract】 Objective:To study the phamacokinetics of three components(berberine hydrochloride, baicalin, jasminodin) in Huanglianjiedu Decoction in rats plasma.Methods: After the rats were administered by Huanglianjiedu Decoction through ig, HPLC was used to determine the three components(berberine hydrochloride, baicalin, jasminodin) in blood plasma at different times, 3P97 procedure was used to process concentration-time data.Results:The whole blood concentration-time course of Berberine hydrochloride and baicalin after oral administration Huanglianjiedu Decoction was fitted to be a one-compartment model, and the same course of jasminodin was fitted to be a two-compartment model. Conclusion:This essay establishes a method for determine three components in Huanglianjiedu Decoction in rats plasma, provided the pharmacokinetic parameters for rational administration of Huanglianjiedu Decoction, and also provided the pharmacokinetic study method of compounds.
