咽拭子标本对手足口病疫情监测的应用研究

【摘要】 [目的]明确致手足口病肠道病毒的主要型别,并对咽拭子标本的核酸检测结果进行评价。[方法]2007年收集青岛市5家医院手足口病患者的咽拭子152份,提取病毒RNA。采用逆转录聚合酶链反应(RT-PCR)方法对病毒RNA进行肠道通用引物及肠道病毒71(EV71)型和柯萨奇病毒16型(CoxA16)特异性引物的检测。[结果]152份咽拭子标本中,83份标本肠道通用引物扩增阳性,阳性率为54.61%。其中,EV71阳性10份,占肠道病毒的12.05%(10/83);CoxA16阳性者42份,占肠道病毒的50.60%(42/83)。[结论]手足口病疫情的病原主要以EV71和CoxA16为主,咽拭子标本在手足口病疫情的监测中具有方便、快捷的优点,而且检出率较高。

【Abstract】 [Objective]To clarify the primary serotypes of causative enteroviruses in hand-foot-mouth disease(HFMD)and to evaluate the role of the throat swab specimens in diagnosis of HFMD.[Methods]152 throat swab specimens were collected from HFMD patients in Qingdao region in 2007.All viral RNA was extracted from the specimens then used as templates in following RT-PCR with enterovirus universal primers.The positive RNA was then tested with two specific primer pairs for VP1 genes of EV71 and CoxA16 respectively.[Results]83 specimens were positive for the universal primers of EV,of which 10 were positive for EV71(12.05%) and 42 for CoxA16(50.60%).[Conclusion]CoxA16 and EV71 were the primary pathogens of HFMD during the epidemic.The throat swab specimen is very suitable in HFMD monitor for its convenient,quick and sensitive characterization.
