海洋真菌多糖YCP多克隆抗体的制备和鉴定

【摘要】 多糖YCP经CDAP活化与牛血清白蛋白(BSA)偶联制备了质量比(WBSA/WYCP)为0.15的拟糖蛋白(YCP-BSA),用该拟糖蛋白免疫新西兰大白兔,制备了抗YCP的抗血清,采用辛酸-硫酸铵法纯化,间接ELISA测定,获得了效价为1∶20 000的YCP多克隆抗体。

【Abstract】 For preparation and characterization of a polyclonal antibody against a new anti-tumor marine polysaccharide,YCP was isolated from marine filamentous fungus Phoma herbarum YS4108,activated by CDAP and subsequently coupled to BSA to form neoglycoprotein.Then rabbits were immunized with the neoglycoprotein.After the fifth immunization,purified anti-YCP antibody titer was quantified by indirect ELISA.Finally,anti-YCP antibody was used for determination of YCP content by indirect ELISA.Neoglycoprotein(WBSA/WYCP=0.15) was prepared and high titer(1∶20000) of antibody was obtained.The detection limit of YCP was 0.5μg/ml by ELISA.Neoglycoprotein prepared by directly linking proteins to CDAP-activated polysaccharides have been shown to induce high titer and protective antibody responses,and the anti-YCP antibody could be applied to detect YCP in blood.
