盐酸氯丙嗪和盐酸异丙嗪制剂的芯片电泳柱后化学发光检测

【摘要】 目的:利用自制整体浇注式 PDMS 电泳芯片,实现药物复方制剂中盐酸氯丙嗪和盐酸异丙嗪的芯片电泳分离和检测。方法:通过柱后化学发光衍生过程,建立微流控芯片化学发光测试方法。结果:两者的分离度可达1.51,盐酸异丙嗪的检出限为2.51×10-8g·mL-1,盐酸氯丙嗪的检出限为5.00×10-8g·mL-1。结论:本方法具有简便快速、分离快速及检测灵敏等优点,显示了该集成微芯片分析系统在药物检测方法学中具有广泛的潜在应用前景。

【Abstract】 Objective:The homemade PDMS CE- chip prepared by PDMS monolithic pouring method was applied for electrophoresis separation with chemiluminescence detection of chlorpromazine hydrochloride and promethazine hydrochloride in the preparation of compounds.Methods:In order to avoid the interference,post column derivation chemiluminescence was adopted.Results:The resolution of chlorpromazine hydrochloride and promethazine hydro- chloride achieved 1.51 with the setup detecting system under optimized conditions,and the detection limit of promethazine hydrochloride and chlorpromazine hydrochloride was found to be 2.51×10-8g·mL-1 and 5.00× 10-8g·mL-1,respectively.Conclusions:The assembled micro -fluidic chip system which can be constructed sim- ply and operated controllably,takes more advantages of rapid separation and detection and high sensitivity,and also demonstrates potential applications in pharmaceutical analysis and clinical diagnose.
