微波萃取法提取当归中阿魏酸的研究
Study on microwave technique for extracting ferulic acid from Radix Angelicae Sinensis
【摘要】 目的:建立测定当归中阿魏酸含量的 HPLC 法。方法:采用 Symmetry C18柱(4.6 mm×150 mm,5μm),流动相为乙腈-0.02 mol·L-1磷酸三氢钾溶液(17:83,以磷酸调节 pH=2.50),流速1.0 mL·min-1,检测波长322 nm,温度:35℃。结果:阿魏酸进样量在0.05~0.13μg范围内具有良好的线性关系(r=0.9998);半均回收率(n=5)为101.1%,RSD 为3.3%。结论:该方法简便、准确,灵敏度高,重复性好。
【Abstract】 Objective:To establish an RP-HPLC for quantitative determination of ferulic acid in Radix Angelicae Sinensis.Methods:Ferulic acid was performed on Symmetry C18 column(4.6 mm×150 mm,5μm)with mobile phase of acetonitrile-0.02 mol·L-1 potassium dihydrogen phosphate buffer solution(17:83,adjusted to pH= 2.50 with phosphoric acid)at a flow rate of 1.0 mL·min-1,detected at 322 nm and column temperature 35℃. Results:The good linear range of ferulic acid was 0.05-0.13μg,r=0.9998.The average recovery(n=5)was 101.1%,and relative standard deviation was 3.3%.Conclusion:This method is testified to be simple,accurate, sensitive and the reproducibility is good.
