复方麝香注射液对氯胺酮麻醉后老年大鼠学习记忆的影响
Effects of Fufang Shexiang injection on dysfunction of learning and memory post-anesthesia with ketamine in aged rats
【摘要】 目的观察老年大鼠氯胺酮麻醉后,学习记忆能力的改变,同时应用复方麝香注射液(SX),以观测其对学习记忆变化的影响。方法28只老年大鼠(20月龄)随机分为4组,每组7只:G1:空白组,第1d(D0)生理盐水(NS)2ml/kg ip,第2d(D1)至第6d(D5)无药物处理;G2:D0:氯胺酮(KTM,50mg/ml)2ml/kg ip,从D1开始每日NS2ml/kg持续至D5;G3:D0:KTM2ml/kg ip,从D1开始每日SX2ml/d持续至D5;G4:D0:KTM2ml/kg ip,从D1开始每日SX6ml/d持续至D5;采用Morris水迷宫仪进行行为学检测。结果随着训练次数的增加,各组逃避潜伏期呈逐渐缩短趋势;G2组:各d逃避潜伏期均长于G1组(p<0.05);G3组:D1至D4与G1组差异无显著性意义,于D3时逃避潜伏期明显短于G2组,D5时明显长于G1组(p<0.05);G4组:各d与G1组比较差异均无显著性意义(p>0.05)。撤除水下平台后游过原平台所在区域次数:G2、G3组游过原平台区次数仍明显低于G1组;而G4与G1组差别无显著性意义(p>0.05)。结论老年大鼠KTM腹腔注射麻醉后,学习记忆能力明显下降,持续至麻醉后5d仍未恢复,SX对此种学习记忆能力下降具有一定改善作用,以较大剂量(6ml·kg-1·d-1)效果显著,较小剂量(2ml·kg-1·d-1)SX对氯胺酮麻醉后的记忆能力下无明显改善效果。
【Abstract】 Objective To observe the effects of Fufang Shexiang injection (SX) on dysfunction of learning and memory post-anesthesia with ketamine (KTM) in aged rats. Methods Twenty-eight SD aged rats (20 months old) were randomized divided into 4 groups (7/group): G1: Conrtol group, at first day (D0), 0.9% Normal sodium (NS) 2ml/kg ip, no drug administration from the second day (D1) to the sixth day (D5); G2: D0 Ketamine (KTM, 50mg/ml) 2ml/kg ip, NS 2ml/kg ip, D0 to D5; G3: D0 KTM 2ml/kg ip, SX 2ml/d, D0 to D5; G4: D0 KTM 2ml/kg ip, SX 6ml/d, D0 to D5. Abilities of learning and memory were observed by Morris Water Maze (MWM). Results Escape latency (EL): The EL showed a tendency that more shorter with more training. G2: EL were longer than G1 all days; G3: At D1 to D4, no significant different with G1, EL shorter than G2 at D3, longer than G1 at D5; G4: No significant different with G1. Times of swimming through the quadrant of the hidden platform after removed it: G1: 4.7 ± 1.8 times; G2 and G3: Less than G1 (p<0.05) ; G4: No significant difference with G1. Conclusions Abilities of learning and memory in aged rats declined after anesthesia with KTM ip, it was proved by MWM test. SX can prevent the decline of learning and memory abilities and large dose(6ml· kg-1· d-1) was more effective. Small dose(2ml· kg-1· d-1) SX had no effects on memory decline.
