【摘要】 目的探讨热休克凋亡的人胃癌细胞致敏的树突状细胞(DC)疫苗对胃癌术后免疫功能的影响。方法从胃癌患者外周血单个核细胞中诱导DC,并用重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)和白细胞介素4(rhIL-4)刺激活化,经热休克凋亡自体胃癌细胞致敏制备DC疫苗。将36例胃癌术后患者随机分为化疗加用DC瘤苗治疗(A)组18例,单纯化疗对照(B)组18例。对两组病例治疗前后T细胞亚群及自然杀伤(NK)细胞及血清IL-2、IL-12、γ干扰素(IFN-γ)水平进行比较。结果A组治疗后外周血CD3+、CD3+CD4+、CD4+/CD8+及NK细胞比率较治疗前明显升高(P<0.05),且明显高于B组(P<0.05);A组治疗后血清IL-2、IL-12、IFN-γ水平较治疗前明显升高(P<0.05)。结论胃癌术后行热休克凋亡自体胃癌细胞致敏的DC疫苗治疗可改善患者的免疫功能。更多还原

【Abstract】 Objective To investigate the effects of dendritic cells(DCs)pulsed with autologous gastric cancer cells lysat on the immune function of the postoperative patients with gastric cancer.Methods DCs isolated from peripheral blood mononuclear cell of gastric cancer patients were cultured and proliferated in vitro using rhGM-CSF and rhll-4,and then were pulsed with autologous gastric cancer cells lysates.Thirty-six postoperative patients with gastric cancer were randomly divided into 2 groups.The dendritic cell vaccine was used in 18 cases(group A),and chemotherapy in 18 cases(group B) as the controls.Immune function and clinical effects were compared between the two groups.Results The levels of CD3+,CD4+,CD4+/CD8+ and NK cell in group A were increased after vaccination(P<0.05),which were not in group B.So did the levels of IL-2,IL-12,IFN-γ in group A and B(P<0.05).Conclusion The gastric cancer cells lysates pulsed DCs vaccines can improve the immune function of the postoperative patients with gastric cancer.更多还原
