放射增敏药马蔺子素对H22肝癌小鼠肺转移的影响

【摘要】 目的探讨放射增敏药马蔺子素对荷瘤动物肺转移的影响及其机制。方法建立小鼠H22肝癌模型,分为健康对照组、肿瘤对照组、单放组、马蔺子素+放组、5-FU+放组。治疗后观察各组肺转移情况;检测T淋巴细胞亚群CD4、CD8;比较各组肿瘤的血管内皮生长因子(VEGF)表达及微血管密度计数(MVD)。结果(1)马蔺子素+放组未发现转移,其余组均有转移;(2)单放组血CD4(27.60±5.04)、CD8(15.94±3.40)较健康对照组(47.74±7.34、22.77±3.46)下降(P<0.01,P<0.05),而马蔺子素+放组CD4(43.3±11.07)回升(P<0.05);(3)马蔺子素+放组肿瘤VEGF评分(2.92±2.02)较肿瘤对照组(4.64±1.02)、单放组(5.86±0.98)、5-FU+放组(4.89±1.78)降低(P<0.01);(4)马蔺子素+放组肿瘤MVD计数(26.14±9.85)较低,单放组(53.29±12.62)较高(P<0.001)。结论在小鼠H22模型上,马蔺子素能抑制肺转移,机制与其提高细胞免疫及减少肿瘤VEGF、MVD表达有关。

【Abstract】 Objective To investigate the impact of irisquinone on lung metastasis in H22-bearing mice and its potential mechanism.Methods Mice were divided into group A(healthy control),group B(tumor control),group C(radiation alone),group D(radiation plus irisquinone)and group E(radiation plus 5-FU).After different treatments,lung metastasis,CD4,CD8,CD4/CD8 level,VEGF score,MVD count of each group were studied.Results No metastasis was found in group D,while it was found in other groups.The levels of CD4 and CD8 in group C were lower than those in group A(P<0.001 or P<0.05).The levels of CD4 and CD4/CD8 in group D were higher than those in group C(P<0.05).Lower expression of VEGF was seen in group D(P<0.01).Lower MVD count was also seen in group D,while higher MVD count was seen in group C(P<0.001).Conclusion For the H22 model,irisquinone can inhibit metastasis,and its mechanism may be related to improving immune function and reduceing the expression of VEGF and MVD in the tumor.
