创伤性休克兔肺组织IL-18、SOD、MDA水平的变化及亚甲蓝的干预作用

【摘要】 目的观察创伤性休克兔肺组织白细胞介素18(interleukin-18,IL-18)、超氧化物歧化酶(superoxide-dismutase,SOD)、丙二醛(malondialdehyde,MDA)水平的变化及鸟苷酸环化酶抑制剂亚甲蓝(methylene blue,MB)的干预作用。方法选用大白兔18只,随机分为对照组(6只)、创伤性休克生理盐水复苏组(生理盐水复苏组,6只)、创伤性休克MB处理组(MB处理组,6只),生理盐水复苏组及MB处理组测定复苏后4h肺组织IL-18、SOD、MDA的水平,对照组测定实验末肺组织IL-18、SOD、MDA的水平。结果兔创伤性休克复苏后肺组织IL-18及MDA明显升高,SOD明显降低,但MB处理组肺组织IL-18及MDA含量明显低于生理盐水复苏组,SOD水平明显高于生理盐水复苏组。结论IL-18及氧自由基在创伤性休克的病理发展过程中起着重要作用,应用MB可降低肺组织IL-18及MDA的水平,增加SOD含量,对创伤性休克缺血/再灌注肺损伤有保护作用。

【Abstract】 Objective To observe the changes of lung tissue interleukin-18(IL-18),superoxidedismutase(SOD)and malondialdehyde(MDA) in rabbit models of traumatic shock and the intervention with methylene blue(MB,a guanylate cyclase inhibitor).Methods Eighteen rabbits were randomly assigned into 3 groups(n=6 each), namely the normal control group,the traumatic shock resuscitated with normal saline group(NS group) and the traumatic shock resuscitated with methylene blue group(MB group).Four hours after resuscitation or by the end of the experiment,lung tissue was taken to measure the contents of IL-18,SOD and MDA.Results In the NS and MB shock groups,the levels of lung tissue IL-18 and MDA were increased and the level of lung tissue SOD was obviously decreased after resuscitation,as compared with that in the control group.But the levels of IL-18 and MDA were obviously lower and the level of SOD was higher in MB group than that in NS group.Conclusions IL-18 and oxygen free radicals play important roles in the pathological process of traumatic shock,and the use of MB in resuscitation can remarkably alleviate the lung injury after traumatic shock by inhibiting IL-18 and MDA production and increasing SOD content.
