银杏叶提取物抗大鼠肺缺血/再灌注损伤的实验研究
Protective effect of extract of Ginkgo biloba against lung ischemia-reperfusion injury in rats
【摘要】 目的探讨银杏叶提取物(EGb761)能否通过诱导血红素氧合酶-1(HO-1)活性减轻肺缺血/再灌注损伤。方法采用呼气末无创血管夹夹闭阻断左主支气管、左肺动脉和左肺静脉30 min后再灌注2 h建立缺血/再灌注(I/R)动物模型。40只雄性Sprague-Dawley大鼠随机分为4组:假手术组、I/R组、EGb组(术前24 h腹腔注射EGb 20 mg/kg)、EGb+ZnPP组(EGb 20 mg/kg+再灌注前15 min静脉注射ZnPPⅨ40μmol/kg),检测肺湿重/干重比、肺泡损伤数、肺组织中HO-1的活性和肺组织中谷胱甘肽过氧化物酶(GSH-Pxase)活性变化。结果①EGb761明显诱导肺组织HO-1活性增加(P<0.01);②同假手术组比较,I/R组肺湿重/干重比及肺泡损伤数增加(P<0.01),肺组织GSH-Pxase活性降低(P<0.01),缺血/再灌注前用EGb761可逆转上述病理改变(P<0.01),用HO-1的抑制剂ZnPPⅨ可部分抑制EGb761的保护作用。结论EGb761通过HO-1的抗氧化作用减轻肺缺血/再灌注损伤。
【Abstract】 Objective To investigate whether the extract of Ginkgo biloba(EGb761) could attenuate the lung ischemia-reperfusion(I/R) injury by inducing the activation of heme oxygenase-1(HO-1) in rats.Methods The lung I/R model was established by ligaturing the left lung hilum for 30 min followed by reperfusion for 120 min in the rats.40 male Sprague-Dawley rats were randomly divided into 4 groups: sham operation group,I/ R group,EGb761 group and EGb+zinc-protoporphyrin-IX(ZnPP Ⅸ) group.The pretreatment with EGb 20 mg/ kg ip and with ZnPP Ⅸ 40 μmol/ kg iv was administered 24 h and 15 min before I/R respectively.The HO-1 activity and GSH-Pxase activity in lung tissue,the lung wet-to-dry weight(W/D) ratio and the number of injured alveoli were determined.Results ①Compared with I/R group,the HO-1 activity was increased markedly in EGb761 group(P<0.01).②Compared with the sham operation group,the W/D ratio and the number of injured alveoli were significantly increased in the I/R group(P<0.01),but the GSHPxase activity was decreased(P<0.01).Pretreatment with EGb761 before I/R could reverse the above changes,likely through the activation of HO-1,which was,in the present study,partially suppressed by ZnPP Ⅸ,an HO-1 inhibitor.Conclusion EGb761 can ameliorate lung I/R injury in rats significantly by activating the HO-1 in lung tissue.
