原花青素和褪黑素对镉致大鼠急性肝氧化损伤影响的实验研究

【摘要】 目的探讨原花青素(PC)和褪黑素(MLT)对镉致大鼠急性肝脏氧化损伤的影响。方法实验用Wistar大鼠32只,按体质量随机分为4组。第1组为对照组,第2组为单纯染镉组,第3组和第4组为PC和MLT预处理组,分别灌胃450 mg/kg PC和5 mg/kg MLT。3 h后第1组皮下注射0.9%生理氯化钠(NaCl),第2、3、4组分别皮下注射35μmol/kg氯化镉(CdCl2),注射容量均为5 ml/kg体质量。染毒24 h后腹主动脉取血并切取肝脏,测定血清中乳酸脱氢酶(LDH)、丙氨酸转氨酶(ALT)活力,测定肝组织中丙二醛(MDA)、谷胱甘肽(GSH)含量及谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活力。结果与对照组比较,单纯染镉组大鼠血清LDH、ALT活力及肝GSH、MDA含量显著升高(分别为257.02、27.13 U/L和10.49μg/g、118.28 nmol/g),GSH-Px、SOD活力明显下降(0.22 U/mg、91.16 U/g)。与第2组比较,PC和MLT预处理组大鼠血清LDH和ALT活力显著降低(分别为181.93、158.15 U/L和12.12、21.08 U/L),肝GSH、MDA含量显著下降(8.74、7.90μg/g和69.99、99.27 nmol/g),GSH-Px、SOD活力明显升高(0.46×103、0.28×103U/g和114.14、106.52 U/g)。结论PC和MLT预处理对镉所致急性肝脏氧化损伤有一定的拮抗作用。

【Abstract】 Objective To study the effects of procyanidins(PC)and melatonin(MLT)on acute hepatic oxidative damage caused by cadmium in rats.Methods Rats were randomly divided into the following four experimental groups(n=8 per group): ① Saline control group;② CdCl2 treated group;③ PC treated group: intragastric administration with 450 mg/kg;④ MLT treated group: intragastric administration with 5 mg/kg.After three hours,the first group was injected subcutaneously with 0.9%NaCl and the other three groups were given CdCl2 35 μmol/kg in total volume of injection was all 5 ml/kg.After twenty-four hours,collected blood from the abdominal aorta and the livers were removed.The activities of LDH and ALT in serum,content of MDA and GSH,and activities of GSH-Px and SOD in the liver were determined.Results Compared with saline control group,CdCl2 treated group serum activities of LDH and ALT increased significantly,levels of GSH and MDA increased significantly and activities of SOD and GSH-Px declined significantly.In contrast,compared to CdCl2 treated group,activities of LDH and ALT declined significantly,the levels of MDA and GSH declined significantly and activities of GSH-Px and SOD in the liver increased significantly in the PC and MLT treated groups.Conclusion Pretreatment with PC and MLT in rats could antagonize acute oxidative liver damage induced by cadmium to a certain extent.
