叠氮化钠对果蝇寿命及某些抗氧化酶活力的影响
Effects of sodium azide (NaN3) on life span and certain activities of antioxidase of Drosophila Melanogaster
【摘要】 目的研究不同浓度的叠氮化钠(NaN3)培养基培养果蝇对果蝇寿命及超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活力的影响。方法用不同浓度(1、4、16、64mg/L)NaN3配制的培养基饲喂野生型黑腹果蝇,并设置基本培养基为对照。寿命实验组每天计数果蝇的死亡数直至全部果蝇死亡,计算果蝇的半数死亡时间、平均寿命与最高寿命。抗氧化酶实验组测定果蝇三龄幼虫SOD及CAT活力,并与对照组相比较进行统计学分析。结果随NaN3浓度的升高,果蝇的平均寿命、半数死亡时间和最高寿命出现明显下降,并呈现剂量-效应关系。在NaN31~16mg/L处理组,随着NaN3浓度的升高,SOD和CAT活力极显著升高,但浓度达到64mg/L,SOD和CAT活力开始下降。结论提示NaN3对果蝇具有一定的毒性效应,并可能会影响其SOD与CAT活力。
【Abstract】 Objective To investigate the effects of sodium azide (NaN3) on life span and the activities of superoxide dismutase (SOD) and catalase (CAT) of Drosophila Melanogaster. Methods Flies were cultured at different concentrations of NaN3(1、4、16、64 mg/L) with culture media as the test group and at basic culture media as the control. In life span groups, dead flies were counted everyday until all flies died, then mean lifespan, half lifespan, maximum lifespan were calculated. In antioxidase groups, the activities of SOD and CAT of the third-instar larvae were determined and compared with the control group. Results With the doses of NaN3 increased, mean lifespan, half lifespan and maximum lifespan were decreased in Drosophila melanogaste. A dose-effect relationship was observed between lifespan and NaN3 groups. SOD and CAT activities in the third-instar larvae were significantly increased at the group of 1-16 mg/L NaN3 in comparison with the control, but decreased obviously at the group of 64 mg/L NaN3. Conclusion It was suggested that NaN3 could have some toxic effects on fruit fly and affect the activities of SOD and CAT of Drosophila melanogaster.
