铅暴露母体的仔鼠脑组织Brn-3a mRNA转录水平变化
Effect of prenatal lead exposure on the gene transcription of Brn-3a POU-domain protein in different regions of rat brain
【摘要】 目的研究铅暴露母体的仔鼠脑组织中POU域蛋白Brn-3a mRNA转录的情况。方法雌性大鼠自妊娠第15天开始经饮水染铅(2.0 g/L,对照组饮蒸馏水)至仔鼠出生后21 d断乳为止。随机取各组仔鼠断头,制作冰冻切片,采用原位杂交检测Brn-3a mRNA水平。结果与对照组相比,大脑皮层和海马部位可见Brn-3a mRNA平均灰度值升高(P<0.05),表明mRNA的表达量减弱,而小脑部位未见明显变化(P>0.05)。海马部位还可见阳性面积比下降有显著性(P<0.01)。结论不同脑区,Brn-3a基因转录水平有不同程度的下降,提示铅可能干扰了Brn-3a的正常表达模式,并由此影响神经元的正常分化。
【Abstract】 Objective To observe the level of gene transcription of POU-domain protein Brn-3a in offspring rat brain after prenatal lead exposure.Methods The pregnant rats were randomly divided into 2 groups: distilled water only or 2.0 g/L lead actate solution in drinking water.The lead-exposure period was limited from the 15th day of pregnancy to the 21th day of giving birth when the offspring began to wean.Brn-3a gene transcription levels were observed by in situ hybridization.Results In situ hybridization showed in some brain regions that mRNA levels of Brn-3a decreased(P<0.05 or P<0.01).Conclusion This study indicated that mRNA levels of Brn-3a decrease to some extent in various brain regions,showing that lead may interfere with the normal expression model of Brn-3a,and so that interferes with the normal differentiation of neuron.
