心肌细胞钙离子的实时动态观察
The dynamic observation of free Ca2+ in cardiac myocytes at low temperature
【摘要】 目的:对SD乳鼠心肌细胞Ca2+在低温条件下进行实时动态观察,为建立低温下心肌细胞钙离子运动模型提供实验依据。方法:采用荧光技术分别于37℃,24℃环境中对分离培养的SD乳鼠心肌细胞胞内Ca2+进行动态观察。结果:心肌细胞在体外呈集落生长。正常条件下,集落中无钙波产生,集落细胞同步搏动;当在24℃的低温环境下,集落中个别细胞先发生钙波,随后集落细胞同步搏动。结论:低温下个别细胞产生的钙波可能对其所在集落细胞的同步搏动有诱导并维持其搏动的作用,这为研究心肌的起搏与传导提供了最简单的模型。
【Abstract】 Objective:To observe the changes of free Ca2+ concentration in cardiac myocytes from SD neonatal rat at low tempera-ture,and to provide references for estabilishing of Ca2+ dynamic movement model at low temperature.Methods:The dynamic movement of free Ca2+ was observed by laser scanning confocal microscope in 37 ℃ and 24 ℃ respectively.Results:The cultured cardiac myocytes in vitro formed colony.In 37℃,cardiac myocytes in the same colony were in synchronous pulsation.In contrast,in 24℃,calcium wave first appeared in some cardiac myocytes of the same colony,then cardiac myocytes in the same colony were beating synchronously.Conclu-sion:The beating of cardiac myocytes was related to the change of Ca2+ concentration.At low temperature,calcium wave happened in some cardiac myocytes may induce and maintain the synchronous beating of cardiac myocytes in the same colony.This work may provide a simple model for cardiac myocytes beating and signal conduction
