心肌活力饮对实验性扩张型心肌病大鼠心肌细胞凋亡及凋亡相关基因的影响

【摘要】 目的观察心肌活力饮对扩张型心肌病大鼠心肌细胞凋亡及凋亡相关基因的影响,探讨该药治疗扩张型心肌病的作用机理。方法60只动物随机分成6组,除正常对照组外,其余各组大鼠分别腹腔注射阿霉素,制造扩张型心肌病模型。各用药组分别予心肌活力饮、黄芪生脉饮灌胃给药。4周后采用流式细胞仪检测心肌细胞凋亡及凋亡相关基因p53和Fas的蛋白表达。结果模型组心肌细胞的凋亡率显著高于正常对照组(P<0.01),凋亡促进基因p53和Fas的蛋白表达增高(P<0.05)。心肌活力饮(5、10、20g原药材/kg)组大鼠心肌细胞相关凋亡基因p53和Fas的蛋白表达量比模型组减少(P<0.05或P<0.01),心肌细胞的凋亡率也明显降低(P<0.01)。结论心肌活力饮可以减少实验性扩张型心肌病大鼠心肌细胞的凋亡,并且可能是通过抑制心肌细胞相关凋亡基因p53和Fas的表达而发挥此作用的。

【Abstract】 Objective To study the effect of Xinji Huoliyin on the apoptosis of myocardial cell and apoptosis-related gene expression of experimental dilated cardiomyopathy rats, and explore its mechanism. Methods Sixty animals were randomly divided into 6 groups. Dilated cardiomyopathy model was made by injecting adriamycin. Experimental rats were given Xinji Huoliyin and Huangqi Shengmaiyin respectively. The genic expression of p53 and Fas was tested by S-P immunohistochemistry technique after 4 weeks. Results The apoptosis rate of myocardial cell of model group was higher than that of normal group (P <0.01), and apoptosis-related gene expression was high in model group (P <0.05). Apoptosis-related gene expression was less in Xinji Huoliyin groups (5, 10, 20 g herbal/kg) than model groups (P <0.05 or P <0.01), and apoptosis rate of myocardial cell in that groups was decresed (P <0.01). Conclusion Xinji Huoliyin can decrese the apoptosis of myocardial cell of experimental dilated cardiomyopathy rats, and may be by inhibiting the expression of apoptosis-related gene.
