反相高效液相色谱法测定湿生扁蕾中木犀草素和1,7-二羟基-3,8-二甲氧酮的含量
Determination of Luteolin and 1,7-dihydroxy-3,8-dimethoxyxanthone in Herbal Gentianopsis paludosa by RP-HPLC
【摘要】 目的建立湿生扁蕾药材中木犀草素和1,7-二羟基-3,8-二甲氧基酮的含量测定方法。方法采用反相高效液相色谱法(RP-HPLC),色谱柱为ZORBAXSB-C18(4.6mm×250mm,5μm);流动相为甲醇(A)-0.4%磷酸水溶液(B)梯度洗脱,0~15min,A∶B=50∶50,15~60min,A∶B=55∶45;检测波长260nm;流速1mL/min;柱温30℃。结果1,7-二羟基-3,8-二甲氧酮和木犀草素分别在0.084~0.84μg和0.184~1.84μg范围内线性关系良好,相关系数分别为0.9994和0.9997;平均回收率分别为96.82%和97.19%,RSD均小于3%。结论该方法简便准确,重现行好,可作为湿生扁蕾药材的质量控制方法。
【Abstract】 Objective To establish the determination of luteolin and 1,7-dihydroxy-3,8-dimethoxyxanthone in Herbal Gentianopsis paludosa. Method The RP-HPLC with ZORBAX SB-C18 (250 mm×4.6 mm, 5 μm) comlum was used. Mobile phase was methanol (A)-0.4% H3PO4 (B) Gradient elution was used, 0~15 min, A∶B=50∶50, 15~60 min, A∶B=55∶45. Detection wavelengh was 260 nm. Flow rate was 1.0 mL/min. Temperature of column was 30 ℃. Result The linear relationship of 1,7-dihydroxy-3,8-dimethoxyxanthone and luteolin were showed at the range of 0.084~0.84 μg and 0.184~1.84 μg (r=0.999 4 and r =0.999 7). The average recovery rate of 1,7-dihydroxy-3,8-dimethoxyxanthone and luteolin were 96.82% and 97.19%, RSD<3%. Conclusion The method was accurate, reproducible and can be used to control the quality of Gentianopsis paludosa.
