丙酮醛诱导细胞凋亡相关基因COX-VA的生物信息学分析
Bioinformatics Analysis for Cell Apoptosis Induced by Methylglyoxal Related Gene COX-VA
【摘要】 在用基因芯片技术检测丙酮醛诱导人牙周膜成纤维细胞凋亡的基因表达研究中,发现一个未知基因表现为显著下调,其名称是COX-VA,并推测该基因与细胞凋亡密切相关。因此利用生物信息学方法对COX-VA基因及其蛋白产物进行各种分析,发现COX-VA基因与细胞色素C氧化酶具有较高的相似性。COX-VA在细胞中低表达与丙酮醛造成线粒体DNA损伤所引发的细胞凋亡相关。
【Abstract】 Previous studies in our lab have shown a down-regulated gene expression(COX-VA) which may be related to apoptosis of human periodontal ligament fibroblasts(HPDLFs),induced by methylglyoxal(MG).In this study,we used bioinformatics analytic methods to further understand the homology,structure of gene and potential functions and location for its coded protein.The results have shown COX-VA has great homology with mitochondrial cytochrome c oxidase.The down expression of COX-VA could lead to mitochondrial DNA damage including oxidative lesions,resulted in apoptosis of HPDLFs induced by MG.
