肉兔AFLP分析体系的建立和优化

【摘要】 以法国伊普吕肉兔的6个品系、新西兰、加利福尼亚兔共8个种群为试材,建立了肉兔扩增片段长度多态性(AFLP)分析体系,对其分析过程中DNA提取、双酶切、连接、预扩增、选择性扩增、银染效果进行了确立。由72种引物组合中筛选出了15种引物组合,建立了肉兔的AFLP指纹图谱。条带清晰可辨,扩增信号强,无背景干扰。

【Abstract】 AFLP(Amplified Fragment Length Polymorphism) analysis system was established with rabbit.The results of several reactions during AFLP analysis were detected such as DNA extraction,double enzymes digestion,adapter ligation,preamplification,selective amplification and silver staining.We Obtained 15 combinations of primers,which had clear and strong amplification bands with bright background.The factors which affected AFLP analysis were discussed.
