pH胁迫下拟穴青蟹体内腺苷三磷酸酶和磷酸酶活性的响应

【摘要】 采用酶学分析的方法研究pH(4.5、6.0、7.5、9.0、10.5)胁迫下拟穴青蟹鳃、肌肉和肝胰腺中ATPase、AKP和ACP活性的响应,以pH6.76±0.3的海水组为对照组.结果表明,随pH胁迫强度加大,拟穴青蟹鳃Na+,K+-ATPase活性显著增强(p<0.05),Ca2+,Mg2+-ATPase活性变化不显著(p>0.05);随pH胁迫时间延长,各组拟穴青蟹鳃Na+,K+-ATPase和Ca2+,Mg2+-ATPase活性均在胁迫第3天出现峰值.肌肉AKP活性对照组显著高于各处理组(p<0.05),pH4.5组肝胰腺AKP活性低于对照组,其余各组均略高于对照组(p>0.05);pH6.0组和pH10.5组肌肉ACP活性显著低于对照组(p<0.05);肝胰腺ACP活性,pH9.0组和pH10.5组显著高于对照组(p<0.05),pH7.5组显著低于对照组(p<0.05),pH4.5组、pH6.0组与对照组差异不显著(p>0.05).随着pH胁迫时间延长,拟穴青蟹肌肉和肝胰腺AKP和ACP活性在相同pH处理组变化规律不明显,但表现出时间效应性.由此可见,pH胁迫显著影响拟穴青蟹的生理生化效应.

【Abstract】 With the method of enzyme analysis,we carried out the experiments for determining the changes of the ATPase and phosphatase activities in gill,muscle and hepatopancreas of Scylla paramamosain under varying pH(4.5,6.0,7.5,9.0 and 10.5) stress for 9 days,with the natural seawater(pH 6.76) as control.The results showed that the activities of Na+,K+-ATPase in gill increased significantly(p<0.05) with pH stress intensity,whereas no marked changes(p>0.05) in Ca2+,Mg2+-ATPase activities were observed,and both Na+,K+-ATPase and Ca2+,Mg2+-ATPase activities reached the peak value at the third day.The AKP activities in muscle were remarkably higher(p<0.05) in control than that of the other groups.Nevertheless,the activities of AKP in hepatopancreas were lower in pH 4.5 group but slight higher(p<0.05) in other groups in comparison with the control group.On the other hand,the muscle ACP activities in both pH 6.0,and pH 10.5 groups were markedly lower(p<0.05) than that of the control group.Compared to the control,the ACP activities of hepatopancreas were significantly higher(p<0.05) in pH9.0 and pH 10.5 groups but lower(p<0.05) in pH 7.5 group.However,no significant difference(p>0.05) was detected among the groups of pH4.5,pH 6.0 and the control groups.The ACP and AKP activities in both muscle and hepatopancreas varied irregularly with the pH stress time,but displayed an obvious time-effect.The present results suggested that pH stress significantly conditioned the physiological-biochemical parameters of Scylla paramamosain.
