IL-18、caspase-1在新生大鼠缺氧缺血脑组织中表达变化及其关系探讨
DEMONSTRATION ON CHANGES OF INTERLEUKIN-18 AND CASPASE-1 PROTEIN AFTER HYPOXIC-ISCHCMIC BRAIN DAMAGE (HIBD) IN NEONATAL RATS
【摘要】 [目的]探讨IL-18和caspase-1在缺氧缺血脑损伤(HIBD)中蛋白质的表达变化,及其二者的关系,进一步揭示HIBD的发病机制,为围产期HIBD的治疗提供新的途径。[方法]7d龄的SD大鼠64只,随机分为:假手术组、HIBD3h、8h、24h、3d、6d、14d组,每组8只。缺氧缺血各组制备左脑HIBD模型,到相应时间点断头取脑采用westernblot的方法测定IL-18和caspase-1蛋白的表达量,比较不同组间蛋白表达的差异。[结果]假手术组、HIBD3h、8h组IL-18蛋白呈低水平表达,在HIBD24h组IL-18蛋白的表达量明显增高,与假手术组相比差异有统计学意义,HIBD14dIL-18蛋白表达量达到高峰(0.8273±0.0675),与其他各组相比P﹤0.01;caspase-1在假手术组、HIBD3h、8h组表达量较低,HIBD24h到6d其蛋白表达呈持续性增强,并于6d达到高峰(0.2839±0.0212,与其他各组相比P﹤0.01),此后仍保持较高水平,但与HIBD6d组相比差异无统计学意义(P﹥0.05)。HIBD后IL-18与Caspase-1蛋白的表达在时间上呈正相关(r=0.8548,P=0.0142)。[结论]新生大鼠HIBD后IL-18、caspase-1蛋白的表达逐渐增加,提示它们均参与HIBD的发病机制。
【Abstract】 [Objective] To investigate the expression of interleukin-18 and caspase-1 protein after hypoxia-ischemia in the neonatal rats’ brain tissue, and further reveal the potential mechanism of hypoxic-ischemic brain damage (HIBD). [Meth- ods] 64 neonatal SD rats, aged seven-day-old and ignored gender, were randomly divided into 7 groups: sham operation group, HIBD3h, 8h, 24h, 3d, 6d, 14d group (n=8 per group) . HIBD model was established by left common carotid artery ligation. The expressions of IL-18 and caspase-1 were analyzed by Western blot. [Results] Lower levels of IL-18 showed in sham operation group and HIBD 3h, 8h group. However, the expression increased in 24h group after HI and compared with sham operation group, there was a significant difference (P﹤0.01). It reached its greatest level at 14d (0.8273±0.0675, P﹤ 0.01 vs. other group) . The expression of caspase-1 in damage brain tissue increased progressively from 24h to 6d after HIBD and reached peak at 6d (0.283 9 ± 0.021 2, P﹤0.01). Although the expression of caspase-1 remained at a high level after HIBD14d (P﹥0.05 vs.HIBD6d group) .The expression patterns between IL-18 and caspase-1 were similar at all time points and there was a positive correlation (r = 0.085 48, P = 0.014 2). [Conclusion] IL-18 and caspase-1 expressions, which in- crease 24h to 14d after HI, most likely happen within the time frame for development of brain injury, and imply that they may play important roles in the pathogenesis of HIBD in neonatal rats.
