HPLC法测定盐酸多西环素片中盐酸多西环素的含量
DETERMINATION OF DOXYCYCLINE HYDROCHLORIDE BY HPLC
【摘要】 [目的]建立HPLC测定盐酸多西环素片的含量。[方法]采用ODS柱(250mm×4.6mm,5μm),以0.05mol/L草酸铵溶液︰二甲基甲酰胺︰0.2mol/L磷酸氢二铵溶液(65︰30︰5)(用氨试液调pH8.3)为流动相,检测波长为280nm。[结果]峰面积对浓度的回归方程为A=21489C-20287,盐酸多西环素在20~120μg/ml浓度范围内,呈良好的线性关系(r=0.9999),平均回收率为100.84%,RSD为0.80%(n=5)。[结论]该法简便、准确,适用于盐酸多西环素片剂的含量的测定。
【Abstract】 [Objective]An HPLC method was established to determine doxycycline hydrochloride.[Methods]The liquid chromatography method was developed,using Alhima C18 column(4.6mm×250mm,10μm),with detection wavelength at 280 nm,mobile phase:ammonium oxalate solution(0.05 mol/L)-N,N-dimethyl formamide-ammonium monohydric phosphate solution(0.2 mol/L )(65︰30︰5). The pH value of the mobile phase is adjusted at 8.3 with ammonia.[Results]The regression equation of peak area and concentration was A=21 489 C-20 287. The linearity is good(r = 0.999 9)in the range of 20~120μg/ml. The recovery rate was 100.84%,RSD=0.80%(n = 5).[Conclusion]The method is simple,accurate and is very suitable for determination of doxycycline hydrochloride .
