腺病毒介导的人CD40配体基因抑制裸鼠卵巢癌生长的实验研究
EXPERIMENTAL STUDY OF ADENOVIRUSMEDIATED HUMAN CD40 LIGAND GENE IN INHIBITING THE GROWTH OF NUDE MICE’S OVARIAN CANCER
【摘要】 [目的]探讨重组腺病毒人CD40配体基因(Ad-hCD40L)对卵巢癌的治疗作用。[方法]建立卵巢癌裸鼠动物模型,检测腺病毒介导的人CD40配体基因对肿瘤生长的抑制作用。[结果]瘤内注射Ad-hCD40L明显抑制裸鼠卵巢癌生长,Tunel法检测和HE染色肿瘤组织石腊切片观察调亡细胞明显增多,并可见大量肿瘤细胞坏死。[结论]腺病毒介导的人CD40配体基因抑制裸鼠肿瘤生长,增强诱导肿瘤细胞调亡,为卵巢癌的基因治疗提供了新的途径。
【Abstract】 [Objective]To investigate the inhibitory effect in treating ovarian cancer with recombinant adenovirusmediated human CD40 ligand gene(Ad-hCD40L).[Methods]A nude mice’s ovarian cancer model was built to exam the anticancer effect of recombinant Ad-hCD40 on the tumor growth.[Results]Human CD40 ligand gene could suppress the growth of tumor in nude mice by intratumoral injection with Ad-hCD40L. Immunohistochemical and HE staining revealed that in Ad-hCD40L treatment group apoptosis obviously increased and a great of tumor cells occurred necrosis comparing with other groups.[Conclusion]The recombinant Ad-hCD40L could suppress the growth of tumor and induce more apoptosis and necrosis in ovarian cancer cells. These findings may provide a new strategy in treating ovarian cancer with gene therapy.
