牙科全瓷修复用长石瓷的生物相容性评价

【摘要】 背景:可加工的长石瓷材料结合计算机辅助设计/计算机辅助加工技术,促进了牙科快速修复技术的进步。修复用全瓷材料不仅应具有合适的机械性能,同时还需要具有良好的生物相容性。目的:通过急性溶血实验、体外细胞毒实验初步评价牙科全瓷修复用长石瓷的生物相容性。设计、时间及地点:单一样本开发性实验,于2004-08/2006-03在首都医科大学附属北京口腔医学院动物实验室完成。材料:长石瓷材料为自制;健康新西兰大白兔1只;小鼠结缔组织成纤维细胞(L-929细胞)为ISO推荐。方法:①急性溶血实验:以长石瓷材料浸提液为实验组、生理盐水为阴性对照组,蒸馏水为阳性对照组3组,分别测定吸光度值,计算被测材料的溶血程度。②体外细胞毒实验:设立100%和50%长石瓷材料浸提液两个浓度的实验组,空白对照组划区分组,每组样本5孔。测定各孔光吸收值,计算细胞增殖率和细胞毒性级。主要观察指标:各组材料的溶血程度及细胞毒性分级。结果:①长石瓷材料的溶血程度值为1.52%,与阴性对照组比较无显著性差异(P>0.05),可认为长石瓷无溶血现象,初步检验合格。②100%和50%长石瓷材料浸提液组A值均与空白对照组无显著性差异(P>0.05),细胞毒性级别均为1级,,满足生物安全要求。结论:自制长石瓷材料的急性溶血实验、体外细胞毒实验结果均满足临床要求,反映其生物相容性初筛实验合格。

【Abstract】 BACKGROUND: Machinable feldspathic ceramic materials with computer-aided design/computer-aided machine technology, can promote the rapid restoration of dentistry. Repair with all-ceramic materials requires not only the appropriate mechanical properties, but also good biocompatibility. OBJECTIVE: To preliminarily evaluate the biocompatibility of feldspathic ceramics to dental all-ceramic materials with in vitro cytotoxicity test and acute haemolysis test. DESIGN, TIME AND SETTING: The single exponent and opening study was carried out in Creatural Testing Center, Faculty of Stomatology, Capital University of Medical Sciences from August 2004 to March 2006. MATERIALS: Feldspathic ceramic materials were self-made; one healthy New Zealand rabbit was used; Mouse connective tissue fibroblasts (L-929 cells) were recommended by ISO. METHODS: ①Acute haemolysis test: Feldspathic ceramic leaching liquor was taken as the experimental group, saline as the negative control group and distilled water as the control group, to detect the absorbance (A) value and calculate haemolysis of the materials.②In vitro cytotoxicity test: Two experimental groups of 100% and 50% feldspathic ceramic leaching liquor, as well as blank control group were set up, 5 holes of samples in each group. A value of each hole was tested to calculate rate of cell proliferation and cytotoxicity level. MAIN OUTCOME MEASURES: Haemolysis and cytotoxicity level of the materials in various groups. RESULTS: ①The haemolysis degree of feldspathic ceramics was 1.52%, and there was no significant difference compared with negative control group (P > 0.05), indicating no haemolysis of feldspathic ceramics that were qualified.②In two experimental groups of 100% and 50% feldspathic ceramic leaching liquor, the A value was similar to that of the control group (P > 0.05), and cytotoxicity was ranked as one, indicating that the material met the biosafety. CONCLUSION: The results of acute haemolysis test and in vitro cytotoxicity test of feldspathic ceramic materials meet clinical requirements, so the biocompatibility of feldspathic ceramics is preliminarily safe for dental application.
