高糖及AGE对高压下THP-1源泡沫细胞ABCA1表达的影响
Influence of high glucose and AGE on ABCA1 expression in THP-1 source foam cell under high pressure
【摘要】 目的观察高糖、AGE及罗格列酮对高压力培养下THP-1源泡沫细胞ABCA1蛋白表达的影响,探讨ABCA1在高血压和糖尿病动脉粥样硬化中的作用机制。方法根据不同处理因素将THP-1源泡沫细胞分为11组:CON组,HP组,HG组,AGE组,HG-HP组,AGE-HP组,HP-R组,HG-R组,AGE-R组,HG-HR组,AGE-HR组。用Westernblot法测定其ABCA1蛋白表达。结果高糖、AGE、高压力及高压与高糖、AGE联合作用明显减弱ABCA1表达(P<0.01),罗格列酮能减弱或取消这种作用。结论THP-1源泡沫细胞ABCA1表达下调可能是高血压及糖尿病动脉粥样硬化的原因之一,其机制可能与PPARγ有关。
【Abstract】 Objective It is to observe the influence of high glucose,AGE and Rosiglitazone on the expression of ABCA1 albumen in THP-1 source foam cell under high pressure,in order to discuss the action mechanism of ABCA1 in hypertension and diabetic atherosclerosis.Methods THP-1 source foam cells were divided into eleven groups including CON group,HP group,HG group,AGE group,HG-HP group,AGE-HP group,HP-R group,HG-R group,AGE-R group,HG-HR group and AGE-HR group according to different preconditioning.The expression of ABCA1 albumen was determined with Westernblot method.Results High glucose,AGE,high pressure,high pressure combined with high glucose and AGE could weaken the expression of ABCA1(P<0.01).Rosiglitazone could weaken or cancel the action.Conclusion The expression lowering of ABCA1 in THP-1 source foam cell maybe one of the causes of hypertension and diabetic atherosclerosis.The mechanism maybe concerned with PPARγ.
