辛伐他汀对骨关节炎模型软骨细胞保护作用的研究
Study on the protective effects of Simvastatin on chondrocytes in osteoarthritis model
【摘要】 目的通过体内和体外实验研究辛伐他汀对骨关节炎模型(OA)膝关节软骨细胞的保护作用。方法取30只6月龄雄性新西兰大白兔,随机分为假手术组(SHAM组,10只)、造模组(ACLT组,10只)和辛伐他汀组(ACLT+SIM组,10只),采用右膝关节前交叉韧带切断术(ACLT)制作骨关节炎模型,ACLT+SIM组每天予辛伐他汀10 mg/kg灌胃。实验持续8周,所有动物在最后一次灌胃的第2天处死。采用酶消化法提取右膝关节软骨细胞进行体外培养,另取SHAM组左侧关节软骨细胞进行IL-1β(10μg/L)诱导的单纯体外关节炎细胞模型制作并予以辛伐他汀(1×10-6mol/L)干扰,分别作为IL组及IL+SIM组。培养2周后提取所有细胞RNA,采用实时荧光定量PCR(Realtime RT-PCR)方法检测骨关节炎发生与保护相关的基因MMP-1、MMP-3、MMP-13a、ggrecan mRNA的表达,同时ELISA法检测细胞上清中及Western Blotting法检测细胞MMP-1的表达,15~19 d CCK-8法检测各组细胞增殖能力。结果体内部分:Aggrecan mRNA的表达在SIM+ACLT组、ACLT组均显著低于SHAM组,余检测mRNA的表达在SIM+ACLT组、ACLT组均显著高于SHAM组;MMP-1的表达在SIM+ACLT组、ACLT组均显著高于SHAM组;细胞增殖的检测结果IL+SIM组与IL组均显著低于SHAM组,余组间无差别。体外部分:IL+SIM组Aggrecan的mRNA表达显著高于IL组,余mRNA的表达在IL+SIM组显著低于IL组(P<0.05),ELISA、Western Blotting检测MMP-1蛋白量与此一致。IL+SIM组与IL组均显著低于SHAM组,但IL+SIM组与IL组无差别。结论口服辛伐他汀不能显著影响兔膝关节不稳定诱发的骨性关节炎模型关节软骨细胞中与骨关节炎发生及保护相关的部分基因的表达,体外给药则可以降低关节炎模型软骨细胞促进骨关节炎发生的相关基因MMPs的表达。辛伐他汀在体内体外均不能刺激软骨细胞的增殖。
【Abstract】 Objective It is to explore the protective effects of simvastatin(SIM) on Chondrocytes of knee joint in osteoarthritis models.Methods 30 male 6 months New Zealand rabbits were divided randomly into three groups: SHAM group(10),ACLT group(10),ACLT+SIM group(10).The rabbits in all the groups except SHAM group were made to osteoarthritis models by anterior cruciate ligament transaction(ACLT) on the right knee.Rabbits in ACLT+SIM group were daily given 10 mg/kg of simvastatin by gavage for 8 weeks.All rabbits were killed on 56th d post-surgery.Chondrocytes isolated from articular cartilage were cultured,and those from left knees of SHAM group were stimulated by IL-1β(10 ng/mL) with(group IL) or without SIM(1×10-6 mol/L)(group IL+SIM).Real-time RT-PCR was used to detect the mRNA expression levels of MMP-1,MMP-3,MMP-13,aggrecan in cultured chondrocytes on the 14th d.MMP-1 contents in the cell culture supernatants were determined by ELISA,and the protein level of MMP-1 in cultured cells were detected by Western Blotting.Cell Counting Kit-8(CCK-8) was used to assay the proliferation of the cultured cells on the 15-19th day.Results The expression levels of mRNA of aggrecan in group ACLT+CT and group ACLT were significantly lower than that in group SHAM,the results of the mRNA expression levels of the other detected genes in group ACLT and group ACLT+SIM were significantly higher than that in group SHAM,so were the results of the protein levels of MMP-1.The results of proliferation ability showed no significant differences within the other three groups.The expression levels of mRNA of aggrecan in group IL were significantly lower than group IL+SIM,the results of the mRNA expression levels of the other detected genes in group IL were significantly higher than group IL+SIM,so were the results of the protein levels of MMP-1.Significantly lower proliferation levels were observed in group IL and group IL+SIM compared with group SHAM.Conclusion A daily simvastatin by oral use at a does of 10 mg/kg for 8 weeks had no significant effects on the expression of genes related to development and prevention of osteoarthritis.But in vitro simvastatin showed chondro-protective effects of simvastatin on osteoarthritis by decreasing the expression levels of MMPs and LEF and increasing the aggrecan expression level.Simvastatin showed no effects on the proliferation of chondrocytes both in vivo and in vitro.
