【摘要】 目的通过观察中药乙肝散的药物血清对肝星状细胞(HSC/T6)的增殖及凋亡的影响,从细胞学和分子生物学水平探讨中药乙肝散的抗纤维化的作用机制,为临床用药提供理论依据。方法①MMT法测定细胞增殖。②用流式细胞仪测定各组各浓度药物血清对HSC/T6作用后的DNA含量,低于G1期DNA含量主峰(亚G1期峰)的细胞为凋亡细胞。结果5%,10%和20%浓度的乙肝散的药物血清均对HSC/T6的增殖有显著的抑制作用,对HSC/T6的凋亡率分别为8.63%,14.56%和21.95%。结论中药乙肝散能抑制HSC/T6的增殖,诱导HSC/T6的凋亡,因此促进活化的HSC的凋亡可能是乙肝散抗肝纤维化的细胞分子学重要机制之一。更多还原

【Abstract】 Objective It is to observe the effect of Yigansan on the proliferation and the apoptosis of HSC/T6,in order to investigate its possible anti-fibrotic mechanism at cellular and molecular lever and provide an effective formula to clinical work.Methods ①MTT assay was used to observe the proliferation of HSC/T6.②Flow cytometry was used to detect the contents of DNA in HSC/T6,The HSC/T6 with sub-G1 peaks in DNA histogram were apoptotic cells.Results The three concentrations as 5%,10% and 20% of Yigansan′s medicated sera could obviously inhibit the proliferation of HSC/T6,the apoptosis rate were 8.63%,14.56% and 21.95%.Conclusion Yigansan can obviously inhibit the proliferation of HSC/T6,induce the apoptosis of HSC/T6.One of the important mechanisms of anti-fibrotic is inducing the apoptosis of activated HSC/T6.更多还原
