姜黄素对慢性应激大鼠肾上腺、血清ACTH和免疫功能的影响
Effects of curcumin on adrenal gland, ACTH in serum and immune function in chronically stressed rats
【摘要】 目的:探讨姜黄素对慢性不可预知应激模型大鼠的肾上腺结构、血清促肾上腺皮质激素(ACTH)浓度和免疫功能的影响及其意义。方法:采用多种不可预知的刺激方式交替、持续应激20d建立大鼠慢性应激模型。随机分为正常对照组、应激模型组、姜黄素组(2.5mg/kg、5mg/kg、10mg/kg)、丙咪嗪组(10mg/kg),每组6只。采用HE染色技术,观察应激大鼠肾上腺结构的改变及姜黄素的作用。通过测定应激大鼠外周血中白细胞(WBC)数量和血清中ACTH和白细胞介素1β(Interleukin-1β,IL-1β)含量的改变,观察姜黄素的作用。结果:慢性应激大鼠与正常对照组比较肾上腺皮质增厚,髓质萎缩,外周血白细胞数量明显减少(包括淋巴细胞、单核细胞和中性粒细胞百分比下降),血清中ACTH和IL-1β含量明显增加。姜黄素组大鼠肾上腺结构变化不明显,外周血WBC数量不同程度增加,血清中ACTH和IL-1β含量降低。结论:姜黄素可能通过调节下丘脑-垂体-肾上腺轴以及免疫系统功能,在慢性应激大鼠模型中显示出抗抑郁活性。
【Abstract】 Objective: To observe the effects of curcumin on the structure of adrenal gland, the level of serum ACTH and immune function in chronically stressed rats. Methods: Male rats were subjected to a variety of unpredictable stressors for 20 days. They were randomly divided into control group, stress group, curcumin group (2.5 mg/kg,5 mg/kg,10 mg/kg) and imipramine group (10 mg/kg). And then regular method was used to prepare pathologic section of adrenal gland, and blood cells analytic system was used to detect white blood cell (WBC) count (including relative WBC percentages) in peripheral blood. The serum level of ACTH and IL-1β were also detected using the enzyme linked immunosorbent assay. Results: Chronic stress resulted in diffuse hyperplasia of the adrenal cortex and atrophy of the adrenal medulla in rats, and the serum level of ACTH was increased, which suggested that stress-adaption failure of the adrenal gland occurred, while adrenal gland of the rats pretreated with curcumin( 10mg/kg) prior to each stressor for 21 days did not result in any pathologic changes and the level of ACTH decreased. Moreover, chronic stress also significantly reduced the WBC count and relative WBC percentages in the peripheral blood, including the percentage of lymphocytes, monocytes, neutrophils, however, curcumin reversed these changes and raised WBC count, along with relative WBC percentages significantly. Meanwhile, the serum level of IL-1βwas increased in chronically stressed animals, and curcumin also declined the level of this stress hormone. Conclusion: These results suggest that curcumin exerts antidepressant effects against chronic stress, and the mechanism may be mediated by regulating the function of the hypothalamo-pituitary-adrenal (HPA) axis and the immune system.
