热休克转录因子1及其突变体的研究进展
Heat shock factor 1 and its mutant
【摘要】 热休克因子1(HSF1)是一种转录因子,在正常时不活化,应激时活化而具有反式激活能力。在其蛋白质结构中含有DNA结合域、三聚结构域、调节结构域、转录活化结构域等,它们在HSF1活化过程中各自发挥重要作用。通过改变HSF1的结构构建其突变体,能特异地引起或阻碍HSF1的活化。这些突变体已应用于心血管疾病、肿瘤、神经系统疾病的研究。
【Abstract】 As a transcription factor,heat shock transcription factor 1(HSF1) is activated during stress and subsequently trans-activate its downstream factors.There are some domains in HSF1 structure such as DNA binding domain,trimerization domain,regulatory domain,and transcriptional activator domain,which play an important effect in process of HSF1 active.Changed structure and prepared HSF1 mutants could specifically active or block HSF1 activation.These mutants had been used to study in various biological questions and will help to overcome some diseases.
