细菌类脂A结构与功能研究进展
Structure and function of lipopolyaccharide lipid A in bacteria——A review
【摘要】 类脂A分布在革兰氏阴性细菌的外表层,它是脂多糖分子的疏水基团。脂多糖,俗名内毒素,可以引起致命的脓毒症、内毒素血症和多器官功能障碍综合症等疾病。近年来的研究发现:脂多糖分子中只有类脂A部分具有内毒素的活性。当细菌侵入人体后,其表面的类脂A可以刺激宿主细胞表面的Toll样受体-4,在细胞内引起一连串的反应,产生一系列的细胞因子。本文根据近年来内毒素领域的国际研究进展,系统综述了类脂A的结构特征、合成途径和致病机理,并在此基础上分析了内毒素在疫苗开发领域的应用前景。
【Abstract】 Lipid A, the hydrophobic group of lipopolysaccharide, covers the surface of most Gram-negative bacteria. Lipopolysaccharide, known as endotoxin, can cause fatal disease like sepsis syndrome. Resent studies have shown that it is only the lipid A part of lipopolysaccharide that has the function of endotoxin. After entering the human body, lipid A on the surface of bacteria can stimulate the Toll-like-receptor 4 on the surface of host cells, cause a series of reaction, and produce different cytokines. Here we have discussed the structure, biosynthesis and pathogenesis of lipid A. The applica-tion of lipid A in vaccine development was proposed.
