【摘要】 目的了解胃癌组织中基质金属蛋白酶-9(MMP-9)基因表达与术前血清中蛋白水平关系,探讨MMP-9表达与胃癌临床病理参数之间相关性。方法45例胃癌组织通过半定量逆转录聚合酶链反应(semi-RT-PCR)、免疫组织化学染色(IHC)、酶联免疫吸附(ELISA)方法检测组织中mRNA及蛋白表达情况,测定术前血清中蛋白水平。结果肿瘤组织基质金属蛋白酶-9mRNA表达程度、蛋白染色阳性率、血清中蛋白水平均显著高于正常对照组(P<0·01);胃癌组织蛋白表达阳性率与肿瘤是否侵犯至浆膜层相关(P<0·05);术前血清中蛋白水平与肿瘤TNM分期、淋巴结转移显著相关(P<0·01)。结论检测基质金属蛋白酶-9表达对胃癌有一定诊断意义;胃癌组织中蛋白表达率及术前血清中蛋白水平对临床肿瘤病情评估具有指导意义;也许多种机制参与了胃癌术前血清中MMP-9蛋白的分泌。更多还原

【Abstract】 Objective To investigate relationship between MMP-9 gene expression and preoperative serum protein level in gastric carcinoma patients,and correlation between MMP-9 expression and pathological features.Methods MMP-9 gene and protein expression in cancer tissue and preoperative serum in 45 cases of gastric carcinoma patients were detected by semi-quantitative RT-PCR,immunohistochemistry,enzyme-linked immunoassay, respectively.ResultsMMP-9 gene expression,protein expression positive rate,preoperative serum protein concentration in gastric cancer group were significantly higher than that in normal control group(P<0.01).Protein expression positive rate was significantly related with cancer invasion degree(P<0.05).Furthermore,preoperative serum protein level in gastric cancer group was significantly related with tumor TNM staging and lymph nodes metastasis(P<0.01).Conclusion Detection of MMP-9 expression is good to gastric carcinoma diagnosis.Evaluation of protein expression and concentration in gastric carcinoma patients are meaningful for gastric cancer progression judgment.Perhaps other mechanisms are involved in MMP-9 protein secretion in preoperative serum of gastric carcinoma,but MMP-9 gene expression and preoperative serum protein level in gastric carcinoma are helpful to diagnosis gastric cancer.更多还原
