【摘要】 目的:探讨Cajal间质细胞在糖尿病胃轻瘫发病机制中的作用。方法:健康雄性SD大鼠20只随机分为正常对照组和糖尿病模型组,每组10只。四氧嘧啶50mg/kg静脉注射1次,建立大鼠糖尿病模型,正常对照组只注射等量生理盐水。营养性半固体米糊法测定胃排空率,免疫组织化学方法和计算机图像分析系统对c-kit阳性的Cajal间质细胞进行定量分析,核酸原位杂交方法和计算机图像分析系统对Cajal间质细胞的c-kitmRNA表达进行定量分析。结果:糖尿病模型组胃排空率及c-kit阳性细胞含量、c-kitmRNA阳性细胞均较正常对照组明显减低(均P<0.05)。结论:糖尿病胃轻瘫大鼠胃Cajal间质细胞的c-kitmRNA表达量减少,导致c-kit含量减少,从而影响Cajal间质细胞正常生物学功能,结构破坏、数量减少,引起胃节律性电活动异常、胃动力下降,胃排空率减低。更多还原

【Abstract】 Objective: To investigate the possible role of interstitial cell of Cajal in the pathogenesis of diabetic gastroparesis. Methods: Twenty healthy male SD rats were randomly divided into 2 groups which included normal control and model group. Diabetes was induced by Ⅳ of alloxan, 50mg/kg except for normal group. Gastric emptying was measured by method of nutritious semisolid paste. C-kit positive cells were quantitively measured with immunhistochemistry assay and computer image analysis system. C-kit mRNA positive cells were quantitively measured with in situ hybridization and computer image analysis system respectively. Results: Gastric emptying rate of the normal group was significantly higher than that of model group(P < 0.05). The number of c-kit and c-kit mRNA positive cells of the normal group were significantly higher than that of model group(P < 0.05). Conclusion: Altered expression of c-kit mRNA may play a key role in the pathogenesis of diabetic gastroparesis.更多还原
