H102对Aβ42致神经元毒性的保护作用

【摘要】 目的:观察H102在细胞水平上对人神经母细胞瘤株SY5Y生长的影响,寻找最佳药物浓度及对Aβ42所致神经元毒性的保护作用。方法:将不同浓度的H102作用于人神经母细胞瘤株SY5Y,利用噻唑蓝(MTT)法测定细胞的存活率,制定浓度-药效曲线得出最佳药物浓度;将人神经母细胞瘤株SY5Y细胞接种后分为对照组、Aβ42损伤模型组及Aβ42损伤加H102保护组,观察各组的MTT代谢率、乳酸脱氢酶(LDH)漏出率、细胞形态。结果:H102在10~160μmol/L浓度范围内均可增加SY5Y细胞的存活率(P<0.01),20μmol/L是增加细胞活性的最适浓度;与对照组相比,Aβ42损伤模型组MTT代谢率下降(P<0.05)、LDH漏出率增高(P<0.01),细胞突起损伤、死细胞增多,加入H102保护后可使上述指标恢复或接近正常。结论:H102具有神经保护作用,可减轻由Aβ42所致的神经元毒性。

【Abstract】 Objective: To investigate the effect of H102 on the growth of human neuroblastoma cell line SY5Y and find out the most effective drug dose and observe the effect of H102 capsule on the neurotoxicity induced by Aβ42. Methods: Different concentrations of H102 were administrated into SY5Y cells. The cell survival rate was measured by MTT. The dose-effect curve was deduced. Human neuroblastoma cells SY5Y were segregated into 3 groups,Aβ42 damaged group,Aβ42+H102 group and control group. MTT metabolic rate,LDH leakage rate and cell morphology were investigated in groups. Results: The SY5Y cell survival rate was increased in 10-160 μmol/L H102 (P < 0.01),the optimal concentration of which was 20 μmol/L. The MTT metabolic rate was decreased in Aβ42 damaged group compared with that of control group(P < 0.05). The elevated leakage rate of LDH,diminished cell axon and increased dead cells were found as well (P < 0.01). All of changes were close to normal after addition of H102. Conclusion: The results suggest the neurotrophic effect of H102 and the diminished effect of neurotoxicity of Aβ42.
