拉莫三嗪对豚鼠娱乐性噪声所致听力损失的保护作用
Protective Effects of Lamotrigine on Noise-induced Hearing Damage in Guinea Pigs
【摘要】 目的探讨拉莫三嗪(lamotrigine,LTG)在豚鼠娱乐性噪声所致听力损失中保护作用的剂量依赖性。方法豚鼠随机分为正常对照组2只,噪声对照组(每天迪厅音乐暴露2h,连续14d)7只,LTG实验组(迪厅音乐暴露2h/d,每天暴露前腹腔注射LTG,连续14d)27只,实验组又分为A组[20mg/(kg.d),i.p.]、B组[30mg/(kg.d),i.p.]、C组[50mg/(kg.d),i.p.],每组各9只。采用听觉脑干诱发电位(auditory brainstem response,ABR)、畸变产物耳声发射(distort productot oacoustic emission,DPOAE)、耳蜗铺片、扫描电镜技术,观察不同剂量LTG对娱乐性噪声所致听阈损失及耳蜗损害的保护作用。结果娱乐性噪声暴露第14天,3个实验组动物较之噪声对照组,有较小听阈偏移幅度及较高DPOAE反应幅值水平,差异有显著性意义(均P<0.01)。其中B、C两组之间无显著性差异,但它们又明显强于实验A组(均P<0.01);3个实验组耳蜗毛细胞琥珀酸脱氢酶(SDH)活性减弱程度比噪声对照组为轻,但差异无显著性意义;扫描电镜示噪声对照组外毛细胞(outer hair cell,OHC)静纤毛异常,而各实验组均接近正常。结论拉莫三嗪能减轻噪声对耳蜗的损害,对听觉功能具有明显的保护作用。
【Abstract】 Objective To investigates whether lamotrigine(LTG) can protect the cochlea from intense noise of disco music-induced hearing damage in guinea pigs.Methods Seven guinea pigs were exposed to discotheque music only for 14 days intermittently(2 h/day)(control group).Twenty-seven of the animals were intraperitoneally injected with different doses of LTG(group A,20 mg/kg every day;group B,30 mg/kg every day;50 mg/kg every day,n=9 in each group) prior to exposure to discotheque music for 14 days intermittently(2 h/day).Dynamic changes of distort product otoacoustic emission(DPOAE) response level and auditory brainstem response(ABR) thresholds evoked by clicks were monitored prior to and during the sound exposure.The sensory epithelium was observed using scanning electric microscopy(SEM) and surface preparation.Results After 14 days of exposure,the auditory thresholds in groups A,B and C were significantly less than those in the control group,whereas the DPOAE level was significantly higher.The SDH activities in hair cells were less diminished in the experimental groups than in the control group,but no significant difference was found among 4 groups.Partial disorders of outer hair cells and stereocilia were only observed in the control group under the scanning electronic microscopy.Conclusion LTG can protect the cochlea against discotheque noise trauma in a dose-dependent manner.
