人体唾液中尿素含量的毛细管电泳/电化学法测定
Determination of Urea in Human Saliva by Capillary Electrophoresis with Electrochemical Detection
【摘要】 首次采用毛细管电泳/电化学法测定了尿素在人体唾液中的含量。考察了工作电极的工作电位、分离电压和进样时间等对分离检测的影响。优化条件下,以直径300μm的铜电极为工作电极,工作电位+0.65 V(vs.SCE),0.25 mol/L氢氧化钠运行液中,尿素质量浓度与峰电流在0.5~2.0 g/L范围内呈现良好线性,检出限为0.05 g/L(0.83 mmol/L)(S/N=3)。该法简单可靠,所需样品很少,对肾病的初步诊断具有一定的参考价值。
【Abstract】 In this work,capillary electrophoresis with electrochemical detection(CE/ED) was utilized for direct measurement of urea in human saliva.This method is based on the high separation efficiency of capillary electrophoresis and the electroactivity of urea at copper electrode in alkaline medium.Saliva samples only needed centrifugation and appropriate dilution prior to analysis.With this method,urea concentrations in saliva samples from both healthy volunteers and renal dysfunction patients were determined,providing useful information for preliminary diagnosis of certain kind of renal diseases.
