龙胆种质资源RAPD评价
RAPD Appraisal of the Germplasm Resources of Gentiana scabra Bge.
【摘要】 目的应用RAPD标记对60份龙胆种质资源遗传多样性进行分析评价,为龙胆种质资源评价与资源库构建提供理论依据。方法应用RAPD对龙胆基因组DNA进行扩增,扩增产物在1.3%琼脂糖凝胶电泳后,在NTSYS-pc软件系统下进行统计分析。结果22条引物共得到147条扩增条带,其中96.6%的片断具有多态性。平均每个引物可获得6.6个DNA片断。各种质间的遗传相似系数(GS)范围为0.235 3 ̄0.821 4,平均值为0.599 2。聚类分析表明:利用RAPD标记将60份供试材料全部分开,根据GS平均值将所有材料分为12类,其中有6份材料分别单独聚为1类,有34份材料聚为1类,其余20份材料分别聚为5类。结论所有材料间的GS值都小于0.83,相似度较低,60份种质间的遗传差异较大,个体间具有较高的遗传多样性水平;RAPD遗传相似系数划分的类群同地理分布有一定关系。
【Abstract】 Objective The genetic diversity of 60 Gentiana scabra Bge.accession was appraised by using RAPD markers in order to afford evidence to germplasm appraising and resource bank construction.Methods Amplification products were separated on 1% agarose after genome DNA of Gentiana scabra Bge.was amplified by using RAPD.The results were statistically analysed with NTSYS-pc.Results A Total of 147 products were amplified by 22 10-mer arbitrary primers,of which 96.6% products were found to be polymorphic.6.6 polymorphic bands were amplified by each polymorphic primers on average.The genetic similarity(GS) between all accessions was 0.235 2~0.824 1.The average GS was 0.599 2 of which.The results of cluster analysis by using UPGMA method showed that all the tested accessions could be differentiated by RAPD markers,and classified as 12 groups,of which 6 accessions was classified as one group separately,and 34 accessions were classified as one group.The rest was classified into 5 groups separately.Conculsions The results also showed that the genetic diversity in Gentiana scabra Bge.based on RAPD.GS was correlated with geographic distribution.Genetic diversity is obvious between accessions.
