不同产地的知母中总皂苷和菝葜皂苷元的含量测定

【摘要】 目的考察不同产地知母中总皂苷、菝葜皂苷元的含量。方法采用香草醛-高氯酸法测定不同产地知母中总皂苷的含量;采用高效液相色谱-蒸发光散射法测定不同产地知母中菝葜皂苷元的含量,色谱柱Waters Nova-Pak C18;流动相为甲醇∶水=90∶10;流速为1.0 ml.min-1;柱温室温,漂移管温度45℃;载气流速30 psi。结果河北、安徽、广东知母中总皂苷的含量为6.04%~7.91%,回收率为99.28%,RSD为2.83%(n=6);菝葜皂苷元的线性范围为1.65~6.6μg(r=0.999 7),河北、安徽、广东知母中的菝葜皂苷元含量为0.72%~0.97%,回收率为106.34%,RSD为2.43%(n=6)。结论各产地知母中菝葜皂苷元含量间变异较大,以河北知母中总皂苷及菝葜皂苷元的含量最高。

【Abstract】 Objective To determine the content of Anemarrhena saponin and sarsa sapogenin in Anemarrhena.Methods The content of Anemarrhena saponin was determined by colorimetry;the content of sarsa sapogenin was determined by HPLC-ELSD.The column was a packed with 5 μm stationary phase Waters Nova-Pak C18 column and a mobile phase of methanol-water(90∶10),with a flow rate of 1.0 ml·min-1,at a temperature of drift tube of 45℃,and the nebulizer nitrogen flow rate was 30 psi,with a universal detector Evaporation Light Scattering Detector(ELSD).Results Anemarrhena saponin in Anemarrhenae was 6.04%~7.91%,and the recovery rate was 99.28 %,RSD<3%(n = 6).The sarsa sapogenin calibration curve was linear in the range of 1.65~6.6 μg(r = 0.999 7) and the recovery rate was 106.34%,RSD was 2.43%(n = 6).Conclusion There are great changes among the different originations.The content of Anemarrhen asaponin and sarsa sapogenin in hebei Anemarrhena is the most.
