高频电火花水针疗法对实验性骨关节炎软骨细胞凋亡的影响

【摘要】 目的研究高频电火花水针疗法对骨关节炎家兔NO及软骨细胞凋亡的影响。方法应用关节制动的方法复制骨关节炎模型,18只造模成功的中国家兔,随机分为高频电火花水针组(简称A组)、水针刀组(简称B组)和对照组(简称C组),A组用高频电火花水针加手法治疗,B组水针刀加手法治疗,C组仅予以手法治疗。1次/d,疗程为1个月,用硝酸还原法检测关节滑液中NO含量;用流式细胞仪检测软骨细胞的凋亡率。结果A、B、C组关节滑液中NO的含量分别为(33.67±17.72)μmol/L、(58.90±22.07)μmol/L,(83.30±16.20)μmol/L,软骨细胞的凋亡率分别为(25.27±10.82)%、(51.71±12.42)%和(74.80±9.37)%。3组间差异有统计学意义。结论与水针刀相比较,高频电火花水针疗法能更显著减少关节液NO的含量,抑制软骨细胞的凋亡,治疗膝骨性关节炎疗效好。

【Abstract】 Objective To study the effects of drug-electric acupotomy(DEA) therapy on nitric oxide(NO) content and articular chondrocyte apoptosis in rabbits with Osteoarthis(OA).Methods The OA rabbit model was established by immobilized in full extension for up to six weeks.Eighteen rabbits were randomly divided into the DEA group(treated with DEA),the acupotomy group(treated with acupotomy and manipulation) and the control group(treated with manipulation alone).The treatment was applied once every three days for 1 month as a course.NO in articular synovia was measured by nitrate reduction method,and the apoptosis rate of chondrocyte was determined by flow cytometry.Results NO content in the A,B,and C groups was(33.67±17.72)μmol/L,(58.90±22.07)μmol/L and(83.30±16.20)μmol/L respectively,and the apoptosis rate in the three groups was(25.27±10.82)%,(51.71±12.42)% and(74.8±9.37)% respectively.There was significant difference among the three groups.Conclusion Compared with the acupotomy plus drug manipulation,DEA plus manipulation could reduce the NO content in synovia fluid and inhibit the apoptosis rate of chondrocytes in treating osteoarthritis of knee joint.
