【摘要】 目的探讨垂体肿瘤转化基因(PTTG)蛋白在急性白血病(AL)中的表达及其与survivin基因表达的相关性。方法应用免疫组化S-P法检测66例急性白血病患者及20例正常对照骨髓单个核细胞PTTG、survivin蛋白的表达,分析其与临床的关系及两者的相关性。结果(1)AL患者BMMNC中PTTG和survivin蛋白表达水平显著高于正常对照组(P<0.01);(2)血清LDH增高组、有髓外浸润组及CR期<6个月组PTTG和survivin表达显著高于LDH正常组、无髓外浸润组及CR期>6个月组,异常染色体核型组PTTG表达高于正常染色体核型组;(3)治疗后达完全缓解(CR)组PTTG和survivin蛋白表达低于未缓解(NR)组;(4)PTTG与survivin蛋白在急性白血病中的表达呈显著正相关(r=0.663,P<0.01)。结论PTTG和survivin基因蛋白在急性白血病骨髓细胞的异常表达可能与白血病的发生、发展有关。更多还原

【Abstract】 Objective To investigate the expression of PTTG and survivin in acute leukemia. Method The expressions of PTTG and survivin protein were examined by immunohistochemical method (S-P) in bone marrow mononuclear cells (BMMNC) of 66 patients with acute leukemia (AL) and 20 normal subjects. Results Expressions of PTTG and survivin in BMMNC of AL patients were significantly higher than those in normal controls (P<0.01). Patients with high LDH, extra-medullary infiltration or short CR phase showed a significantly higher expression of PTTG and survivin than those with low LDH, no extra-medullary infiltration or long CR phase. The higher expression of PTTG was showed in abnormal karyotype groups compared with normal karyotype group. Expressions of PTTG and survivin were lower in AL patients who reached CR than those in patients whose chemotherapy failed. Expression of PTTG was positively correlated with that of survivin(r=0.663, P<0.01). Conclusion The overexpression of PTTG and survivin may be involved in the progression of leukemia.更多还原
