【摘要】 目的:探讨提高巴氏涂片质量的改良方法。方法:尝试应用冷冻式巴氏涂片技术对宫颈病变进行检测,观察改良后巴氏涂片的形态效果。结果:涂片的细胞量充足,重叠减少、形态结构清晰,无皱缩自溶现象,着色良好,色彩艳丽,浆核对比突出,胞质、核染色质、核仁和其他细胞成分易于识别,细胞境界清楚,透明度良好。结论:冷冻式巴氏涂片技术是建立在传统巴氏涂片基础上的一种改良技术,该法在制片的过程中强调注重取材制片的全过程,其核心步骤是把固定好后的细胞玻片置入细胞形态修复液中,利用渗透膨胀的原理对已干枯萎的细胞进行修复,从而达到恢复细胞形态的目的,促进细胞着色,提高细胞透明度和清晰度,有利于对病变细胞的识别和提高对宫颈病变细胞的阳性检出率、特异度和灵敏度,降低漏诊误诊率。该技术无需特殊设备及耗材,成本低廉,操作简单,实用性强,具有较好的应用推广价值。我们还认为,传统巴氏涂片的缺点不是固有不变的,而是取决于临床及细胞病理医生之间的互动,他们是否真正具备耐心和以谨慎的态度来重视巴氏涂片的各个细节是提高巴氏涂片质量的关键。更多还原

【Abstract】 Objective To investigate on the method of improving papanicolaou test quality.Methods Attempt to use freezing technology of papanicolaou test for detecting cervical lesion and observing cervical cells morphology changes under light microscopes.Results An adequately cellular cervical cytology smear,the reduce of cells overlay,the integrality of cells morphology,the clear structure of cells and unconspicuous phenomenon of dissolving and cell shrinkage were observed,the cells were stained well and flamboyantly,the contrast ration was outstanding between cytoplasm and nucleu,the cytoplasm,nucleus,karyosome,and other components were easy to be resolved,the cells boundary,transparence showed clearly.Conclusion The freezing technology of papanicolaou test is one meliorative technique based on papanicolaou test,the technique emphasizes on every process of sampling,smearing and staining,the most important step is the fixed smears should be soaked into the restoring-liquid of cells morphology based on the theory of penetration and expansion principle,thus achieve the purpose to cells restoring their normal appearance,to promoting cells staining and to improving cells transparency and clarity,it will be favorable to improve the cells recognition rate,the positive rate,the specification,sensitivity and decrease leaking rate and misdiagnosis rate.The meliorative technique is simple and low cost,it needn’t special equipments and consumables,it has rather good practicability and reliability that would have a great future in spread and application.We also think that the shortcomings of traditional papanicolaou test are not constant but depend on the interaction between clinician and cytopathologist,their patience and attitude are the key for High Quality of papanicolaou test.更多还原
