HPLC法测定不同产地返魂草中绿原酸的含量
Determination of contents of chlorogenic acid in Senecio cannabifolius Less. from different regions by HPLC
【摘要】 目的建立同时测定不同产地返魂草中绿原酸含量的方法。方法采用HPLC法。色谱柱为Diamonsil ODS C18柱(4.6 mm×250 mm,5μm),流动相为乙晴-体积分数为0.4%的磷酸水溶液,检测波长为326 nm;流速为1 mL.min-1。结果绿原酸质量在62~301 ng内与峰面积呈良好的线性关系,平均回收率为99.2%,RSD为0.91%(n=9);5个产地返魂草中绿原酸含量分别为0.95、0.720、.55、0.64、0.44 mg.g-1。结论HPLC法可用于返魂草中绿原酸的含量测定;5个产地的返魂草中绿原酸含量有明显差异,吉林通化返魂草中绿原酸含量最高。
【Abstract】 Objective To set up HPLC method for the determination of the contents of chlorogenic acid in Senecio cannabifolius Less.from different regions.Methods A Diamonsil C18 column(4.6 mm×250 mm,5 μm)was used with the mobile phase consisting of acetonitril and 0.4% phosphoric acid.The flow rate was 1 mL·min-1 and the detection wavelength was set at 326 nm.Results The liner range of chlorogenic acid was 62-301 ng.The average recovery of chlorogenic acid was 99.2%(RSD=0.91%,n=9).Contents of chlorogenic acid were 0.95,0.72,0.55,0.64,0.44 mg·g-1 from five regions respectively.Conclusions The method is suitable for determination of chlorogenic acid in Senecio cannabifolius Less.There exist significant differences in the contents of chlorogenic acid in Senecio cannabifolius Less.from five regions.Among them,the content of chlorogenic acid from Jilin Tonghua is the highest.
