人白细胞介素24重组蛋白对胃癌抗肿瘤机制的实验研究
Study on Anti-tumor Mechanisms of Recombinant Human Interleukin 24 Protein
【摘要】 目的纯化人白细胞介素24(IL-24)的原核重组表达质粒pET-21a(+)-IL-24在大肠杆菌BL(DE3)中的表达产物rhIL-24,并初步探讨其对胃癌细胞株SGC-7901抗肿瘤的可能机制。方法采用RT-PCR法和Western blot法检测rhIL-24对胃癌细胞株SGC-7901中促凋亡因子bax表达的影响,鸡胚绒毛尿囊膜技术观察其对血管形成的影响。结果rhIL-24处理组胃癌细胞中促凋亡因子bax表达增高,rhIL-24处理组二、三级血管数目明显减少,三级血管的数目与生理盐水组比较,差异有统计学意义(P<0.05)。结论初步表明rhIL-24能上调促凋亡因子bax表达,从而抑制血管生成,发挥对胃癌的抗肿瘤作用。
【Abstract】 Objective To purify the expression product of procaryon recombinant expression plasmid of human IL-24 in E coli, and investigate the anti-tumor mechanisms of rhIL-24 on hu- man gastric cancer cell line SGC-7901. Methods RT-PCR,Western blot were used to detect the expression of bax in human gastric cancer cells line SGC-7901, and observe the effect of rhIL-24 on chorioallantoic membrane. Results In human gastric cancer cells line SGC -7901 treated by rhIL-24, the expression of bax was increased significantly. The number of blood vessel was de- creased and it had significant difference when compared with the control group (NS). Conclusion rhIL-24 may inhibit the grow of gastric cancer by promoting the expression of bax and restraining angiogenesis.
