阿霉素对心肌细胞损伤的实验研究
Research on the Damage of Myocardial Cells induced by Adriamycin
【摘要】 目的研究阿霉素对体外培养乳鼠心肌细胞的损伤作用。方法采用MTT法观察阿霉素对乳鼠心肌细胞活性的影响;通过检测乳酸脱氢酶(LDH)释放量,观察阿霉素作用下的乳鼠心肌细胞的坏死程度;采用流式细胞仪检测阿霉素诱发的心肌细胞凋亡百分率。结果(1)MTT实验发现:阿霉素浓度在0.3~10.0μg/ml均能抑制心肌细胞活性并成剂量依赖性;(2)细胞心肌酶谱检测到LDH释放量与阿霉素浓度呈正相关;(3)流式细胞仪发现心肌细胞凋亡率与阿霉素浓度有相关性。结论一定浓度的阿霉素对心肌细胞有毒性作用,可致心肌细胞损伤,表现为心肌细胞坏死和凋亡发生率增加。
【Abstract】 Objective To study the damage induced by adriamycin(ADM) in cultured murine myocardial cells.Methods After exposed to adriamycin,the viability of cardiac muscle cells was assessed by MTT colorimetric assay and apoptosis rate was determined by flow cytometry,the lactate dehydrogenase(LDH) in myocardial was examined by blood biochemical autoanalyzer.Results(1)Adriamycin inhibited the growth of rat cardiac muscle cells and had a dose-dependment or time-dependment manner in a certain range of dose.(2) The relationship between release of lactate dehydrogenase(LDH) in myocardial and the dose of adriamycin was positive and time-dependment.(3) Apoptosis rate assessed by flow cytometry was correlated with adriamycin.Conclusion Adriamycin has toxic effect,causing the damage of cardiac cells or apoptosis.
