前列腺素A1预处理供体大鼠对心脏移植受体T细胞功能的影响

【摘要】 目的研究核转录因子-кB(NF-кB)抑制剂前列腺素A1(PGA1)预处理供体大鼠,在同种心脏移植中对供心的保护及对其受体T淋巴细胞功能影响。方法使用大鼠同种心脏移植排斥模型,观察PGA1预处理供体对心脏存活时间影响,并进行受体大鼠混合淋巴细胞培养及测定血清白介素2、白介素4(IL-2,IL-4)浓度。结果PGA1组供心存活时间较生理盐水组延长(P<0.05),混合淋巴细胞培养显示PGA1组受体大鼠T细胞对供体抗原刺激的增殖反应较生理盐水组受到抑制(P<0.05),PGA1组血清IL-2浓度较生理盐水组降低(P<0.05),而两组间血清IL-4浓度无显著差异(P>0.05)。结论PGA1预处理供体能有效延长供心存活时间,抑制受体T淋巴细胞的免疫功能。

【Abstract】 Objective To observe protective effects of the NF-кB inhibitor PGA1 on donor heart and to study effects of PGA1 on the function of recipient T-cells in rat model of heart transplantation.Methods Donor were pretreated with PGA1 then the isotype dystopy rat heart transplantation was performed.Mean survival time of donor heart was determined.Six rats in each group was sacrificed on the 5th day after transplantation and then mixed lymphocyte culture(MLC) and levels of serum IL-2、IL-4 were determined.Results The survival time of cardiac allograft in PGA1 group were prolonged than that in the normal solution group(P<0.05).MLC showed that proliferative responses of spleen cells to alloantigen stimulation from PGA1 group of recipients were suppressed(P<0.05),the serum IL-2 level was reduced in the PGA1 group(P<0.05),and there were no difference in the level of serum IL-4 between two groups(P>0.05).Conclusion PGA1 can inhibit graft rejection and prolong survival time of cardiac allografts.
