生防木霉菌T41菌株生物学特性研究
Biological Characteristics of Trichoderma Strain T41
【摘要】 木霉菌T41菌株是一株分离于土壤中的具有生防潜力的木霉菌株,为进一步研究菌株的发酵培养及田间应用技术提供理论依据,进行了菌株生物学特性研究。采用室内菌丝生长、产孢测定方法研究了T41菌株生物学特性,结果表明:T41菌株在PDA、PSA、PMA、NA、查彼培养基和基本培养基上均能生长,但在PDA培养基上菌丝生长和产孢最佳,菌落直径达到81mm,产孢量达到2.6×1010个;温度对菌丝生长和孢子产生影响显著,在10~40℃范围能够生长产孢,以30℃时菌丝生长最快,培养2d后菌落直径达到93mm,并且产孢量也高于其他培养温度;光照处理对菌丝生长影响不明显,但光照可明显促进孢子产生。葡萄糖、木糖、果糖和麦芽糖能够较好促进菌丝生长,葡萄糖和果糖是T41菌株产孢的最佳碳源;蛋白胨、天冬酰胺是有利于菌丝生长和孢子产生的有机氮源;微量元素中Ca、Cu对菌丝生长及产孢都有一定的促进作用,而Mn、Zn则不利于T41菌丝生长和产孢;菌株在pH值2~11的范围均可以生长产孢,但偏酸性条件是T41菌株生长产孢的适宜环境,pH值5为最佳。
【Abstract】 Trichoderma T41 is a potential biocontrol fungus that was isolated from soil.Though it could grow on PSA,PMA,NA,Czapek and basic cultrure medium,PDA is the most favorable medium for the hypha growth and spore production.Temperature had distinct effect on the hypha growth and sporulation,from 10℃to 40℃ it could grow and produce spore,but the optimal temperature for the growth and sporulation of T41 was 30℃.For the basic culture medium,the glucose,xylose,fructose and maltose was optimal carbon source for hypha growth,glucose and fructose were propitious to spore production.Protein peptone was the best nitrogen source for hypha growth,protein peptone and asparagin were the best nitrogen source for spore production.The pH for hyphae growth and spore production ranged from 2 to 11,the optimal pH was 5.
