DNA修复基因XRCC1单核苷酸多态性与铂类药物化疗对外周血白细胞的影响相关性分析
The Relationship between Polymorphisms of DNA X-ray Repair Cross Complementary Protein 1 and the Changes of White Blood Cell after Platin-Based Chemotherapy
【摘要】 目的探讨X射线损伤修复交叉互补基因1单核苷酸多态性与应用铂类药物化疗前后外周血白细胞变化的关系。方法化疗前采集外周静脉血,用多聚酶链反应—高温连接酶反应(PCR-LDR)进行XRCC1Arg399Gln基因多态性的分型;观察化疗前后白细胞计数改变的情况。结果XRCC1Arg399Gln各基因型与化疗后白细胞总数、中性粒细胞计数下降无相关性(P>0.05);携带XRCC1399Arg/Gln及Gln/Gln基因型的患者化疗后淋巴细胞计数低于Arg/Arg者,而淋巴细胞下降的相对数要高于Arg/Arg者,化疗淋巴细胞下降绝对值无显著性差异。结论XRCC1399Arg/Gln基因型与化疗后淋巴细胞下降相关,在一定程度上可以预测化疗后淋巴细胞减少的程度。
【Abstract】 Objective To investigate the relationship between genetic polymorphisms of DNA X-ray repair cross complementary protein 1(XRCC1) and the changes of white blood cell(WBC) after platin-based chemotherapy.Methods Peripheral venous blood was used to determine the polymorphisms of XRCC1 codon Arg399Gln by polymerase chain reaction and ligase detection reaction(PCR-LDR).WBC counts were performed before and after chemotherapy.Results No significant difference of the WBC and neutrophil counts after chemotherapy was discovered between the XRCC1 codon Arg399Gln genotypes.The lymphocyte counts in the cases with 399Arg/Gln or Gln/Gln was significantly lower than that with Arg/Arg after chemotherapy.The relative number of lymphocyte decrease in the cases with 399Arg/Gln or Gln/Gln was significantly more than that with Arg/Arg,but there was no significant difference in the absolute number of decrease of lymphocyte among the XRCC1 codon Arg399Gln genotypes.Conclusion Polymorphisms of XRCC1 Arg399Gln might be associated with the decrease of lymphocyte after platin-based chemotherapy and could be useful to predict the degree of lymphocyte decrease.
